吡柔比星与丝裂霉素C对膀胱癌T24细胞株抑制作用的实验研究

摘    要:目的探讨吡柔比星(THP)与丝裂霉素C(MMC)对膀胱癌T24细胞株的抑制作用。方法将膀胱癌T24细胞株分为4组,分别给予THP、MMC、THP+MMC和MMC+THP。用MTT法测定T24细胞的增殖抑制率,用TuNEL法检测T24细胞的凋亡情况。结果THP、MMC、THP+MMC和MMC+THP四组的增殖抑制率(%)分别为54.3±6.4,48.9±7.2,62.3±5.9,69.1±6.2;凋亡指数(%)分别为6.2±2.5,4.4±2.7,6.7±3.0,7.4±3.2。联合用药组对T24细胞的抑制作用优于单药THP和MMC对肿瘤细胞的抑制作用(P〈0.05)。在联合用药组中,MMC+THP组对T24细胞的抑制作用优于THP+MMC组对肿瘤细胞的抑制作用(P〈O.05)。结论THP和MMC均可抑制膀胱癌T24细胞株的增殖并诱导凋亡,两药序贯联合用于膀胱灌注化疗可能具有合理性。

Abstract:Objective To investigate the inhibiting effect of pirarubicin and mitomycin C on the bladder cancer cell line T24. Methods The inhibitory effects of THP, MMC, THP+MMC, MMC+THP on bladder cancer cell line T24 were determined with MTT assay. The apoptosis indexes were detected with terminal deoxynucleotide transferase mediated dUTP-nick end labeling method. Results The inhibition rates of THP, MMC, THP+MMC, MMC+THP were (54.3±6.4)%, (48.9 ±7.2) %, (62.3± 5.9) % and (69.1± 6.2) % respectively, and the apoptosis indexes were (6.2±2.5) %, (4.4± 2.7) %, (6.7 ±3.0)% and (7.4±3.2)% respectively. The inhibitory effect was found when THP and MMC were used alone or together, with better inhibitory effects when THP and MMC used together (P〈0.05). MMC+THP showed better effect than THP+ MMC(P〈0. 05). Conclusions THP and MMC showed significant inhibiting effect on bladder cancer cell line T24, which indicated that sequential administration of THP and MMC might have satisfactory clinical effect for bladder cancer patients.
