Talin-1与原发性肝癌的侵袭和迁移

摘    要:目的探讨Talin与不同来源人肝癌细胞侵袭及迁移的关系,进而为进一步研究Talin与肝癌的侵袭和迁移的可能机制提供理论基础。方法体外培养人高转移肝癌细胞株(MHCC-97H细胞株)、人低转移肝癌细胞株(MHCC-97L细胞株)、人肝癌SMMC-7721细胞株、人肝癌Be L-7402细胞株、人肝癌He PG-2细胞株及人正常肝脏细胞株(LO2细胞株),RT-PCR技术检测各细胞株中Talin-1的mRNA表达水平,Western-blot技术检测各细胞株中Talin-1的蛋白表达水平,Transwell侵袭和迁移实验及细胞划痕实验检测各细胞株侵袭及迁移能力,软琼脂集落形成实验检测各细胞株增殖和克隆能力。结果五种人肝癌细胞株中Talin-1的mRNA表达水平不同(P0.05),五种人肝癌细胞株中Talin-1蛋白表达水平不同,MHCC-97L细胞株中Talin-1的蛋白表达比LO2细胞株高(P0.05);五种人肝癌细胞株的侵袭能力不同(P0.05),五种人肝癌细胞株的迁移能力不同(P=0.0000),与正常人肝脏细胞相比,五种人肝癌细胞株的增值和克隆能力不同(P=0.00017)结论 Talin-1与肝癌的侵袭和迁移能力相关;高表达Talin-1的原发性肝癌的侵袭和迁移能力低,恶性度低,低表达Talin-1的原发性肝癌的侵袭和迁移强,恶性度高,Talin-1可作为原发性肝癌恶性度的预测指标之一。

Talin-1 is correlated with invasion and migration in Primary liver cancer

Abstract:Obstract To study the level of Talin-1 expression is correlated with invasion and migration in human hepatocellular carcinoma. Methods We cell culture the highly metastatic human liver cancer cell line MHCC-97 H cell line and the minimally metastatic liver cancer cell lines MHCC-97 L cell line and human liver cancer lines such as MHCC-97 L cell line,MHCC-97 H cell line,SMCC-7721 cell line,BEL-7402 cell line,He PG-2 cell line and normal human liver cell(LO2 cell line),We measured the mRNA and protein expression levels of Talin-1 in five human liver cancer cell lines and normal human liver cell(LO2 cell line) by real-time PCR and western blot. The migration and invasion of the cells were assessed using transwell assays and cell scratch experiments,respectively,and proliferation was assessed by soft AGAR colony formation. Results Talin-1 mRNA expression differed significantly between liver cancer cell lines and the normal liver cell line(LO2 cell line)(P〈0. 0001). The protein expression of Talin-1 in the liver cancer cell lines and the normal liver cell line differed significantly. MHCC-97 L cell line exhibited elevated Talin-1 protein levels(P〈0. 05). Compared with the LO2 cell line,the invasion and migration abilities of the five cancer cell lines differed significantly(P〈0. 05). The colony-forming ability differed among the cancer cell lines compared with the LO2 cell line(P = 0. 00017). Conclusion Talin-1 is associated with the invasion and migration of human liver cell lines. High levels of Talin-1 expression are correlated with reduced invasion and migration as well as decreased malignancy in human liver cancer cell lines; the suppression of Talin-1 promotes invasion and migration. Talin-1 can serve as a one of predictors of primary liver cancer in malignant degree.
