白介素-6在自发性高血压大鼠肾损害中的表达和意义

目的 研究白介素-6在自发性高血压大鼠肾组织的表达及其意义.方法 以同龄雄性正常血压大鼠和自发性高血压大鼠为研究对象,检测10周龄和28周龄两种大鼠尾动脉压、24 h尿蛋白定量、尿β2微球蛋白、肾功能；取肾组织作苏木精-伊红染色、免疫组织化学及反转录-聚合酶链式反应法检测白介素-6蛋白及信使核糖核酸表达情况.结果 与同龄正常血压大鼠比较,自发性高血压大鼠尾动脉压、24 h尿蛋白定量和尿β2微球蛋白明显增高,28周龄自发性高血压大鼠较10周龄明显增高.肾组织白介素-6蛋白及信使核糖核酸表达自发性高血压组较同龄正常血压大鼠增强,28周龄自发性高血压大鼠较10周龄自发性高血压大鼠增强,白介素-6表达与24 h尿蛋白定量、尿β2微球蛋白呈正相关.结论 白介素-6在自发性高血压肾组织表达增强,可能是高血压肾损害发生的重要机制之一.

Expression and significance of interleukin-6 in renal damage of rats with spontaneous hypertension

Objective To study the expression of interleukin-6 in the renal tissues of rats with spontaneous hypertension. Methods The arteria eaudilis pressure, renal funetion,24-h urinary protein and urinary β2-mieroglobulin in rats with spontaneous hypertension and Wistar-Kyoto rats were meas- ured at the 10 th and 28 th week. The renal tissue specimens were taken for hematoxylin-eosin staining and the expression levels of interleukin-6 in the renal tissue were examined by immunohistochemistry and reverse tanseription-polymerase chain reaction. Results Compared with the same age Wistar-Kyo- to rats,arteria eaudilis pressure, 24-h urinary protein and urinary ~2-microglobulin in rats with sponta- neous hypertension were significantly increased. In rat with spontaneous hypertension, the arteria eaud- ilis pressure, 24-h urinary protein and urinary ~32-microglobulin at 28 th week were significantly higher than those at 10 th week. The expression of interleukin-6 mRNA and protein in the renal tissues of rats with spontaneous hypertension was significantly increased as compared with the same age Wistar-Kyo- to rats. The expression of interleukin-6 mRNA and protein in the renal tissue of rats with spontaneous hypertension at the 28 th week was significantly increased as compared with those at the 10 th week. The expression of interleukin-6 was positively correlated with 24-h urinary protein and urinary mi- croglobulin- Conclusions The expression of interleukim6 in the renal tissue is increased in rats with spontaneous hypertension,which is involved in the hypertensive renal damage.
