细胞膜片技术及其在骨组织领域的应用进展

摘    要:目的对细胞膜片技术(cell sheet technology,CST)及其在骨组织领域的应用进展进行综述。方法广泛查阅近年来国内外相关文献,并进行总结。结果 CST主要应用温度反应培养皿收获细胞,避免使用蛋白酶,保留了培养过程中细胞自分泌的细胞外基质及相关蛋白和因子,将细胞以一层完整的膜状结构收集,为体内骨愈合提供了适宜的微环境。以CST为基础,通过同型或异型细胞膜片叠加、细胞膜片结合支架以及细胞膜片折叠等方法构建三维结构组织,在组织工程骨构建方面表现出了巨大潜力。结论 CST与传统组织工程方法结合将促进骨组织工程学的发展。

Abstract:Objective To review the progress of cell sheet technology(CST) and its application in bonetissue engineering.Methods The literature concerning CST and its application was extensively reviewed andanalyzed.Results CST using temperature-responsive culture dishes is applied to avoid the shortcomings of traditionaltissue engineering.All cultured cells are harvested as intact sheets along with their deposited extracellular matrix.Avoiding theuse of proteolytic enzymes,cell sheet composed of the cells and extracellular matrix derived from the cells,and remained therelative protein and biological activity factors.Consequently,cell sheet can provide a suitable microenvironment for the boneregeneration in vivo.With CST,cell sheet engineering is allowed for tissue regeneration by the creation of three-dimensionalstructures via the layering of individual cell sheets,be created by wrapping scaff old with cell sheets,or be created by folding thecell sheets,showing great potential in tissue engineered bone.Conclusion Constructing tissue engineered bone using CSTand traditional method of bone tissue engineering will promote the development of the bone tissue engineering.
