脂蛋白(a)对鼠腹腔巨噬细胞的细胞毒性作用

摘    要:目的探讨脂蛋白(a)对巨噬细胞释放一氧化氮的影响,进一步了解脂蛋白(a)在动脉粥样硬化发生发展中的作用。方法将脂蛋白(a)(终浓度为2.5~40mg/L)与鼠腹腔巨噬细胞共孵,检测上清液中乳酸脱氢酶活性、一氧化氮含量和细胞一氧化氮合酶活性。结果在基础及脂多糖诱导状态下,细胞上清液中一氧化氮(NO)含量与脂蛋白(a)呈显著的剂量依赖性降低,并呈时间效应。而细胞一氧化氮合酶活性则不受脂蛋白(a)刺激的影响。结论脂蛋白(a)对巨噬细胞释放NO有明显抑制作用。

Abstract:Objective To explore the effect of lipoprotein (a) on macrophages releasing nitric oxide, in order to further investigate the role of lipoprotein (a) in the development of atherosclerosis. Methods Lipoprotein (a) (final concentrations ranged from 2.5 to 40 mg/L) was coincubated with murine peritoneal macrophages. After lipopolysacharride stimulation for 48 h, lactate dehydrogenase activity and nitric oxide concentration in the cultured supernatant as well as cellular nitricoxide synthase (NOS) activity were measured. Results Under both basic and stimulated conditions, nitric oxide concentration in the cultured supernatant decreased in lipoprotein (a) dose dependent manner and the effect was time-dependent. While, cellular NOS activity didn't change under lipoprotein (a) stimulation. Conclusion Lipoprotein (a) significantly inhibits releasing of nitric oxide from macrophages.
