光纤DNA生物传感器的应用特征

摘    要:目的:总结近年来国内外有关光纤DNA传感器应用的研究近况,探讨光学DNA传感器在基因检测中的应用。 资料来源:应用计算机检索Medline1990—01/2005—12相关光纤DNA传感器应用的文献,检索词“DNA biosensor,optical fiber”,并限定文献语言种类为English。同时计算机检索CNKI1990-01/2005—12相关光纤DNA传感器方面的文献,检索词为“传感器,DNA,光纤”,并限定语言种类为中文。 资料选择:对资料进行初审,选取包括光纤DNA传感器的文献,开始查找全文。纳入标准:光纤DNA传感器在基因检测中的应用:排出标准:①电化学,压电晶体DNA传感器的应用。②综述文献、重复研究、Meta分析类文章。 资料提炼:共收集到35篇关于光纤DNA传感器的文献,纳入21篇符合标准的文献,10篇文献对光纤DNA传感器的应用进行评价。 资料综合:光纤DNA生物传感器是近年来发展很快的一项新技术,是分子生物学、分析化学、光电子学的交叉技术。光纤DNA传感器对基因的检测具有灵敏度高、特异性好,易于排出非特异性因素的干扰。光纤DNA传感器可检测杂交后产生的特异性信号,选择性强,易于排除杂交过程中非特异性吸附干扰。可应用于模式寡核苷酸的检测、DNA损伤的检测、基因诊断检测、致病菌的检测、药物检测、对酶的检测。 结论:光纤DNA传感器基因检测方面的应用是完全可行的,但还需进一步改进,使其能够对肿瘤、遗传病等进行检测。

Abstract:OBJECTIVE: To summarize the latest application researches of optical fiber DNA biosensor in recent years, and explore the application of optical DNA biosensor in gene detecting.
DATA SOURCES: The related literatures of optical fiber DNA biosensor were searched in the Medline and CNKI database during January 1990 and December 2005 by using the key words of "DNA, biosensor, optical fiber".
STUDY SELECTION: After the primary inspection, the literatures containing the optical fiber DNA biosensor were selected for full-texts. Inclusive criteria: The application of optical fiber DNA biosensor in gene detecting. Exclusive criteria: (1) the application of electrochemical, piezoelectric crystal DNA biosensor; (2)reviews, repeated study or Meta- analytical papers.
DATA EXTRACTION: Totally 35 literatures about optical fiber DNA biosensor were collected, and 21 papers were in accordance with the inclusive criteria while 10 papers were used to evaluate the application of optical fiber DNA biosensor.
DATA SYNTHESIS: Optical fiber DNA biosensor is a new developing technology in recent years, and also a crossing technology including molecular biology, analysis chemistry and optoelectronics. Owing to its high sensibility, good specificity and easy ejection for the obstruction of non-specific factors. optical fiber DNA biosensor is effective for not only detecting the specific signals produced by cross, but also detecting pattern oligonucleotide, DNA damage, gene diagnosis, pathogenic germ, drug and enzyme.
CONCLUSION: It is feasible to detect genes by using optical fiber DNA biosensor. However, the deep researches will be carried out to apply it on the detections of cancer and hereditary disease.
