宫颈癌组织YAP1和SRSF3蛋白表达与临床病理参数及预后的关系

摘    要:目的检测宫颈癌组织Yes相关蛋白1(Yes-associated protein 1, YAP1)、丝氨酸/精氨酸富集剪接因子3(serine and arginine rich splicing factor 3, SRSF3)蛋白表达情况,探讨YAP1、SRSF3蛋白表达与临床病理参数及预后的关系。方法 81例宫颈癌患者,取其宫颈癌组织和癌旁正常组织,采用免疫组织化学法检测宫颈癌组织和癌旁正常组织YAP1和SRSF3蛋白表达情况。记录患者临床资料,分析YAP1、SRSF3蛋白表达与临床病理参数的关系;采用Pearson相关性分析宫颈癌组织YAP1与SRSF3表达的相关性;采用Kaplan-Meier生存曲线分析YAP1、SRSF3蛋白阳性表达者与阴性表达者3年生存率的差异;采用多因素COX比例风险回归模型分析宫颈癌患者预后不良的影响因素。结果宫颈癌组织YAP1、SRSF3蛋白阳性表达率(74.07%、65.43%)高于癌旁正常组织(28.40%、30.86%)(P<0.05)。TNM分期Ⅰa/Ⅰb、无淋巴结转移的宫颈癌患者YAP1(65.31%、64.58%)和SRSF3(51.02%、52.08%)阳性表达率均低于TNM分期Ⅱa、有淋巴结转移患者(87.50%、87.88%,87.50%、84.85%)(P<0.05)。宫颈癌组织YAP1与SRSF3表达呈正相关(r=0.708,P=0.001)。YAP1阴性表达者3年生存率(90.5%)高于YAP1阳性表达者(70.0%)(P<0.05),SRSF3阴性表达者3年生存率(92.9%)高于SRSF3阳性表达者(66.0%)(P<0.05)。TNM分期Ⅱa期(OR=2.036,95%CI:1.024~3.049,P<0.001)、淋巴结转移(OR=1.842,95%CI:1.569~2.216,P=0.006)、YAP1表达阳性(OR=1.480,95%CI:1.203~2.429,P<0.001)和SRSF3表达阳性(OR=1.525,95%CI:1.106~1.814,P=0.011)是宫颈癌患者预后不良的危险因素。结论宫颈癌组织YAP1和SRSF3阳性表达率明显提高,YAP1和SRSF3表达与TNM分期、淋巴结转移、预后和生存相关,在宫颈癌的预后和诊断中有一定临床价值。

Abstract:Objective To detect the expressions of Yes-associated protein 1(YAP1) and serine/arginine rich splicing factor 3(SRSF3) in cervical cancer tissues, and to explore the correlations of the expressions of YAP1 and SRSF3 proteins with the clinicopathological parameters and prognosis. Methods The expressions of YAP1 and SRSF3 proteins in cervical cancer and adjacent normal tissues from 81 cases of cervical cancer were detected by immunohistochemistry. The clinical data were recorded, and the correlations of the expressions of YAP1 and SRSF3 proteins with the clinicopathological parameters were analyzed. Pearson correlation analysis was used to evaluate the correlation of YAP1 with SRSF2 in patients with cervical cancer. Kaplan-Meier survival curve was used to analyze the difference of 3-year survival rate between positive and negative expressions of YAP1 and SRSF3 proteins. COX proportional hazard regression was used to analyze the influencing factors of the poor prognosis of cervical cancer patients. Results The positive rates of YAP1 and SRSF3 proteins were higher in cervical cancer tissues(74.07%, 65.43%) than those in adjacent normal tissues(28.40%, 30.86%)(P<0.05). The positive rats of YAP1(65.31%, 64.58%) and SRSF3(51.02%,52.08%)in patients with TNM stageⅠa/Ⅰb and no lymph node metastasis were lower than those in patients with TNM stageⅡa and lymph node metastasis(YAP1:87.50%,87.88%;SRSF3:87.50%,84.85%)(P<0.05).YAP1 was positively correlated with SRSF3 in cervical cancer tissues(r=0.708,P=0.001).The 3-year survival rate was higher in patients with negative expression of YAP1(90.5%)than that in patients with positive expression of YAP1(70.0%)(P<0.05),and higher in patients with negative expression of SRSF3(92.9%)than that in patients with positive expression of SRSF3(66.0%)(P<0.05).TNM stageⅡa(OR=2.036,95%CI:1.024-3.049,P<0.001),lymph node metastasis(OR=1.842,95%CI:1.569-2.216,P=0.006),positive expression of YAP1(OR=1.480,95%CI:1.203-2.429,P<0.001)and positive expression of SRSF3(OR=1.525,95%CI:1.106-1.814,P=0.011)were the risk factors of poor prognosis of cervical cancer patients.Conclusion The positive rates of YAP1 and SRSF3 in cervical cancer tissues are significantly elevated.The expressions of YAP1 and SRSF3 are correlated with TNM stage,lymph node metastasis,prognosis and survival,and both of them have certain clinical value to the prognosis and diagnosis of cervical cancer.
