实验性脑出血大鼠急性期血浆内皮素和降钙素基因相关肽含量变化的研究

摘    要:目的:探讨血浆内皮素(ET)和降钙素基因相关肽(CGRP)在实验性脑出血大鼠急性期中的变化规律,初步分析其在脑出血急性期中的病理生理意义。方法:用Nath 法复制脑出血大鼠模型,放射免疫法动态测定急性期血浆ET及CGRP含量,电镜下观察脑组织超微结构变化。结果:实验组血浆ET 含量在各观察点均明显高于对照组(4 小时P< 0.05,12 小时与7 日P均< 0.01,24 小时与72 小时P均< 0.001),7 日时间点实验组与正常组比较差异非常显著(P< 0.01)。实验组血浆CGRP含量在各时间点亦明显高于对照组(4 小时及7 日P 均< 0.05,12、24 和72 小时P均< 0.01),7 日时间点与正常组比较有显著性差异(P< 0.05)。结论:血浆ET 及CGRP在脑出血急性期均升高,其变化规律基本一致,与脑损伤程度呈正相关,提示二者对脑出血急性期的病理演变过程有重要意义

Abstract:Objective:To investigate the pathophysiological significance of changes in plasma endothelin (ET) and calcitonin generelated peptide (CGRP) levels in rats with experimental intracerebral hemorrhage during acute period.Methods:A rat model of acute intracerebral hemorrhage was made by Nath method.ET and CGRP levels in plasma were dynamically measured by radioimmunoassay and ultrastructure changes in brain were also observed.Results:ET levels in the experimental group were significantly higher than that in the control group at various time points (at 4 hours, P <0 05;at 12 hours and 7 days,both P <0 01;at 24 and 72 hours,both P <0 001);ET levels in the experimental group at 7 days were also markedly higher compared to the normal group ( P <0 01).Similarly,plasma CGRP levels were obviously higher than that in the controls during the observation period (at 4 hours and 7 days ;both P <0 05;at 12,24 and 72 hours,all P <0 01),and it was significantly higher at 7 days compared to the normal group ( P <0 05).Conclusions:Both plasma ET and CGRP levels elevated during acute intracerebral hemorrhage and they were positively correlated with the degree of brain damage.These data suggest that ET and CGRP may play an important role in the pathological response to acute intracerebral hemorrhage.
