螺内酯对 2 型糖尿病大鼠血脂及凝血四项的影响

摘    要:目的 探讨非选择性醛固酮受体拮抗剂-螺内酯(SPI)对2型糖尿病大鼠(T2DM)血脂及凝血四项的影响.方法 高脂高糖喂养加小剂量链脲佐菌素(STZ)腹腔注射法建立2型糖尿病大鼠模型,将成膜的T2DM大鼠随机分为糖尿病对照组(DM-C组)和SPI干预组(DM-S组),继续高脂高糖喂养,正常对照组(NC组)大鼠继续给予普通饲料喂养,其中DM-S组给予SPI 40 mg/(kg·d)灌胃,NC组和DM-C组灌胃等量蒸馏水.每两周监测大鼠体重和血糖水平,第16周末处死大鼠,收集血液标本,监测空腹血糖(FBG)、糖化血红蛋白(HbA1c)、甘油三酯(TG)、血钾(K+);凝血四项:凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB).HE染色观察16周末大鼠血管内皮病理学变化.结果 DM-C组和DM-S组大鼠的FBG、HbA1c%、TG均高于NC组(均P<0.01),而DM-S组和DM-C组比较差异无统计学意义;3组之间的血钾水平差异无统计学意义(P>0.05);DM-C组和DM-S组大鼠PT、APTT均低于NC组(P<0.05),FIB则高于NC组(P<0.05),而DM-C组和DM-S组之间无显著差异(P>0.05).结论 螺内酯对糖尿病大鼠血糖、血脂及凝血无明显改善,但可以减轻血管内皮的损伤程度.

Abstract:Objective To investigate the effect of nonselective aldosterone antagonist receptor - spimnolactone (SPI) on the coagulation and the Vascular endothelial of type 2 diabetic mellitus (T2DM) rats. Methods The model of T2DM rats was established by fed with high - su-crese -high -fat diet and injected low dose streptozotocin (STZ). These models will become the model of the rats. All these models were random- ly divided into two groups : diabetes mellitus control group ( group DM - C) and diabetic rats were treated with spironolactone ( group DM - S). Each rat in group DM - C and DM - S were fed with the high - sucrose - high - fat diet. Normal control rats ( group NC) were fed with routine an- imal feeds. The group DM - S was treated with spironolactone 40 mg/( kg · d) via intragastric administration. The group DM - C and NC were treated with corresponding distilled water. Body mass (BM) and peripheral blood glucose of each group were tested every two weeks. The levels of blood glucose (FBG) , glycosylated hemoglobin Alc (HbAlc) , triglyceride (TG) and four coagulation were tested at the end of 16th week. HE staining was used to observe the pathological changed of vascular endothelial after 16 weeks. But there was no difference between group DM - C and DM - S ( P 〉 O. 05 ). Results The levels of blood glucose ( FBG), HbAlc, TC, TG and FIB at the 16th week in group DM - C and group DM -S were higher compared with group NC ( P 〈0.01). The level of PT and APTT in group DM - C and DM - S was lower compared with group NC ; but there was no difference between group DM - C and DM - S ( P 〉 O. 05 ). Conclusion SPI may play the protective role for vascu- lar endothelial in diabetic rats, but not affect the serum and coagulation.
