钛表面单宁酸络合氯化钙涂层接枝氯己定功能及稳定性探究

摘    要:目的探究钛表面单宁酸和氯化钙络合后接枝氯己定的生物相容性、抗菌性以及稳定性。以期在种植体表面构建持久抗菌涂层。方法商业钛片经打磨后,以光滑钛片为对照组,接枝不同浓度的氯己定。通过X射线光电子能谱分析元素组成。接触角测量仪分析其亲水性,Zeta电位实验分析其反应稳定性,金黄色葡萄球菌的涂板计数法,抑菌环法以及增值法分析其抗菌性能。甲基噻唑基四唑法和荧光显微镜观察细胞培养1d,3d的细胞活性,以及浸泡1、4、7、14、30、60d后测量涂层的稳定性。结果单宁酸和钙络合后钛表面C、Ca原子百分含量明显增加,接触角下降,zeta电位数值下降;接枝氯己定后,接触角上升,菌液涂板未见菌落,抑菌环透亮,细菌增值OD为0.06;细胞培养1、3 d氯己定组细胞活性与对照组无显著性差异(P>0.05),氯己定组钛表面黏附大量细胞,且细胞形态良好。浸泡1、4、7、14、30、60天后抑菌环,细菌涂板法以及增值法分析其有良好的抗菌性。结论钛表面单宁酸和氯化钙络合后接枝氯己定有良好的生物相容性和优良的抗菌抑菌能力,且具有持久的稳定性。

Study on the Function and Stability of Grafting Chlorhexidine onto Tannic Acid Complexing Calcium Chloride Coating on Titanium Surface

Abstract:Objective To investigate the biocompatibility,antibacterial activity and stability of the grafted chlorhexidine after complexation of tannic acid and calcium chloride on titanium surface.To construct long-lasting antibacterial coating on implant surface.Methods Commercial titanium plate after grinding,smooth titanium plate as control group,grafted with different concentrations of chlorhexidine.The composition of the elements was analyzed by X-ray photoelectron spectroscopy.The hydrophilicity was analyzed by contact Angle measuring instrument,the reaction stability was analyzed by Zeta potential test,and the antibacterial properties were analyzed by staphylococcus aureus smear counting method,bacteriostatic ring method and increment method.Methylthiazolyl tetrazolium method and fluorescence microscope were used to observe the cell activity of 1d and 3d culture,and the stability of the coating was measured after soaking for 1,4,7,14,30 and 60 days.Results After the complexation of tannic acid and calcium,the content of C and Ca atoms on the titanium surface increased obviously,the contact Angle decreased,and the Zeta value decreased and became stable.After the grafting of chlorhexidine,the contact Angle increased,no colonies were found on the bacterial liquid coating plate,the inhibition ring was transparent,and the bacterial proliferation OD was 0.06.There was no significant difference in cell activity between the chlorhexidine group and the control group at 1 and 3 days of cell culture (P>0.05),a large number of cells were attached to the surface of titanium in chlorhexidine group,and the cell morphology was good.After soaking for 1,4,7,14,30,60 days,the antibacterial ring,bacterial coating method and increment method were used to analyze the antibacterial activity of the product.Conclusions The grafting of chlorhexidine onto tannic acid and calcium chloride on titanium surface has good biocompatibility and excellent antibacterial and bacteriostatic ability,and has lasting stability.
