肝癌患者外周血单个核细胞HLA-G表达研究

目的了解肝细胞癌（HCC）患者外周血单个核细胞（PBMC）人类白细胞抗原G（HLA—G）的表达水平。方法收集38例HCC患者、25例肝硬化患者和40名健康查体者的PBMC，分别用逆转录一聚合酶链反应（PCR）和流式细胞术（FCM）检测其HLA—GmRNA和HLA—G蛋白表达水平。结果HCC组、肝硬化组和健康对照组PBMCHLA—GmRNA表达率分别为94．7％、84．0％和87．5％，差异无统计学意义（P=0．359）；HCC组HLA—G蛋白表达率高于肝硬化组和健康对照组，差异有统计学意义（P=0．002）。HCC组PBMCHLA—G蛋白主要表达于CD4^＋细胞上。结论HCC患者PBMCHLA—G蛋白表达率升高，其机制与临床意义尚待进一步研究。

Research on the expression of peripheral blood mononuclear cell HLA-G in patients with hepatocellular carcinoma

Objective To investigate the expression level of peripheral blood mononuclear cell （PBMC） human leucocyte antigen G （HLA-G） in patients with hepatocellular carcinoma （HCC）. Methods The HLA-G mRNA and HLA-G protein expression levels on PBMC from 38 patients with HCC, 25 patients with fiver cirrhosis and 40 healthy subjects were determined by reverse transeription-polymerase chain reaction （PCR） and flow cytometry （FCM）. Results The HLA-G mRNA expression rates in HCC group, cirrhosis group and healthy control group were 94.7% ,84.0% and 87.5% , respectively,and there was no statistical significance （P = 0. 359）. The ttLA-G protein expression rate in HCC group was significantly higher than those in cirrhosis and healthy control groups with statistical significance （ P = 0.002）. In HCC group, the HLA-G protein was mainly expressed on CIM ＋ cells. Conclusions The PBMC HLA-G protein expression rate in HCC patients increases, and its mechanism and clinical significance need to be further researched.
