摘 要: | 目的 探讨高浓度葡萄糖对3T3 L1脂肪细胞脂联素分泌的影响。 方法3T3 L1脂肪细胞诱导分化后分为4组,处理如下:(1)5.5mmol/L葡萄糖;(2)5.5mmol/L葡萄糖+50μmol/L PD98059;(3)17.5mmol/L葡萄糖;(4)17.5 mmol/L葡萄糖+50μmol/LPD98059。48小时后半定量RT PCR法检测3T3 L1脂肪细胞PPAR γmRNA的表达,ELISA法检测培养液中脂联素浓度。 结果 (1)高浓度葡萄糖可以降低PPAR γmRNA的表达、减少3T3 L1脂肪细胞脂联素的分泌;(2)PD98059能够部分逆转由高糖引起的PPAR γmRNA的表达的降低及脂联素分泌的下降。 结论 高浓度葡萄糖通过激活MAPK信号传导通路降低PPAR γmRNA的表达,同时减少3T3 L1脂肪细胞脂联素的分泌。

The effect of high glucose on the secretion of adiponectin in 3T3 L1 adipocytes |

Abstract: | Objective: To study The effect of high glucose on the secretion of adiponectin in 3T3 L1 adipocytes. Methods: After 3T3 L1 adipocytes have been differentiation, they were divided into four groups and were treated as follows: (1) 5.5mmol/L glucose; (2) 5.5mmol/L glucose+50μmol/L PD98059;(3) 17.5 mmol/L glucose; (4) 17.5 mmol/L glucose+50μmol/L PD98059. After being cultured for 48 hours, the expression of PPAR γmRNA was measured by semi quantitative RT PCR with β actin as internal control and adiponectin in cluture solution were detected by ELISA methods. Result: (1) High glucose could reduce the expression of PPAR γ mRNA and decrease the secretion of adiponectin in 3T3 L1 adipocytes.(2)PD98059 could partially reverse the inhibitory effect of high glucose on the PPAR γ mRNA and decrease the secretion of adiponectin in 3T3 L1 adipocytes expression of PPAR γmRNA and the secretion of adiponectin. Conclusion: High glucose could reduce the expression of PPAR γmRNA and decrease the secretion of adiponectin in 3T3 L1 adipocytes via the activation of mitogen activated protein kinase (MAPK) pathway. |
