摘    要:目的:研究电针(EA)结合经颅磁刺激(rTMS)对急性脑缺血大鼠不同脑区神经生长因子(NGF)、脑源性神经营养因子(BDNF)及mRNA的表达。方法:120只成年雄性Wistar大鼠随机分成正常组、模型组、EA组、rTMS组和EA+rTMS组(分别为A、B、C、D、E组)各24只。除A组外,其它组均复制急性大脑中动脉缺血模型,选取1d作为开始治疗的时间点,C、D组和E组分别施以EA、rTMS和EA结合rTMS方法处理。在治疗后的7、14及28d3个时间点各组分别取8只,采用免疫组织化学检测法、Westernblot以及RealtimePCR法检测大鼠脑组织NGF、BDNF及mRNA的表达。结果:C、D组和E组大鼠梗死灶周围以及海马区NGF、BDNF及mRNA的表达增强(P〈0.05),尤其以E组表现更明显,28d后各组差异无统计学意义。结论:EA结合rTMS治疗能上调NGF、BDNF及mRNA的表达。

Abstract:Objective:To study the effect of electroacupuncture (EA) combined with transcranial magnetic stimulation (rTMS) on the expression of NGF and BDNF in different cerebral regions of rats with cerebral ischemia. Methods: 120 male Wistar rats were randomly divided into 5 groups: normal group, model group, EAgroup, rTMS group and EA+ rTMS group. After the establishment of acute middle cerebral artery occlusion (MCAO) , the rats in various groups were accordingly treated with EA, rTMS or EA plus rTMS, respectively. Immunohistochemistry, Western blot and realtime PCR were used to detect the expression of NGF and BDNF. Results: In EA, rTMS, and EA plus rTMS groups, the expression of NGF, BDNF was increased at the 7th, and 14th day as compared with the model group (P〈0.05), especially in the EA plus rTMS group. Conclusion: EA plus rTMS can promote the expression of NGF, BDNF.
