通脑精胶囊对胰岛素抵抗大鼠局灶性脑缺血后脑组织自由基的影响

摘    要:目的 研究通脑精胶囊对胰岛素抵抗 (IR)大鼠急性脑梗死IR状态、脑组织超氧化物歧化酶(SOD)活性和脑组织丙二醛 (MDA)含量的影响 ,进一步探讨其作用机制。方法 通过高糖饲料喂养复制IR大鼠模型 ,在此基础上采用电凝大脑中动脉造成局灶性脑缺血 ,观察通脑精胶囊对血清胰岛素 (Ins)、胰岛素敏感指数 (ISI)、脑组织SOD活性及MDA含量等指标的变化。结果 与空白对照组比较 ,IR基础上的局灶性脑缺血大鼠血清Ins水平显著升高 (P <0 0 1) ,ISI明显降低 (P <0 0 1) ,脑组织SOD活性显著降低 (P <0 0 1)及脑组织MDA含量明显增高 (P <0 0 1) ;经通脑精胶囊治疗后 ,上述指标均有明显好转 (P <0 0 1)。结论 IR大鼠局灶性脑缺血后存在明显的IR状态 ,脑组织SOD活性显著降低 ,MDA含量明显增高 ;通脑精胶囊具有逆转IR、增强脑组织SOD活性、降低脑组织MDA含量的作用 ,从而达到脑保护的作用 ,推测这可能是该方改善急性脑梗死的作用机制之一。

Abstract:Objective To investigate the effect of Tongnaojing capsule(TNJC) on the insulin resistance(IR),the activity of superoxide dismatase(SOD) and the contents of malonaldehyde(MDA) of cerebral tissue in rats with IR after local cerebral ischemia,and its mechanism.Methods The models of rats with IR were established by feeding the high-carbohydrate forage,then local cerebral ischemia was produced by the electrocoagulation of the middle cerebral artery.And the effect of the TNJC on the changes of serum insulin(Ins),insulin sensitivity indexes(ISI),the activity of SOD and the contents of MDA of cerebral tissue were observed.Results Compared with the control group,the serum Ins in the model group was markedly increased(P<0.01),ISI significantly decreased(P<0.01),the activity of SOD decreased markedly(P<0.01) and the contents of MDA increased significantly(P<0.01).Indexes above improved significantly after TNJC administration(P<0.01).Conclusion There were the obvious IR state,the decrease of the activity of SOD and the increase of the contents of MDA in rats with IR after local cerebral inshemia.The TNJC has the effect of reversing IR,strengthening the activity of SOD and decreasing the contents of MDA of brain tissue,which might be one of the mechanisms in the treatment of acute cerebral infarction by the TNJC.
