2型糖尿病患者外周血中膜型及可溶性CD28分子的表达及意义

摘    要:目的分析2型糖尿病患者外周血中膜型及可溶性CD28分子的表达及其意义。方法分别收集2012年12月至2013年12月在北京军区联勤部门诊部就诊的30例2型糖尿病患者(糖尿病组)及同期30名健康体检者(对照组)的外周血,通过溶血及流式细胞术检测其外周血T细胞表面CD28分子的表达,酶联免疫吸附(ELISA)法检测血清中可溶性CD28的水平,并进行对比分析。结果糖尿病组外周血CD4+CD28+T细胞和CD8+CD28+T细胞显著低于对照组(28.29±7.71)%比(32.77±4.28)%和(12.18±3.17)%比(18.53±3.27)%,均P0.01],血清中可溶性CD28水平显著高于对照组(2.86±0.96)比(0.61±0.19)μg/L,P0.01]。结论协同刺激分子CD28很可能参与了2型糖尿病的免疫病理机制。

Abstract:Objective To analyze the expression of membrane type CD28 and soluble CD28 in peripheral blood of type 2 diabetes patients. Methods The peripheral blood mononuclear cells( PBMCs) of 30 subjects with type 2 diabetes( diabetes group) and 30 healthy donors( control group) were collected from December 2012 to December 2013 in the out- patient department of Outreach Logistics Department of Beijing Military Region. The expression of CD28 on T cells in peripheral blood of type 2 diabetes patients was detected by flow cytometry( FCM). The secretion of soluble CD28 in serum was detected using the skills of Enzyme- linked immunosorbent assay( ELISA).The data was analyzed by the Graphpad Prism 5. 01 program. Results The percentage of CD4^ + CD28^ + and CD8^+ CD28 ^+ T cells in peripheral blood of type 2 diabetes patients were( 28. 29 ± 7. 71) % and( 12. 18 ± 3. 17) % respectively,and were decreased markedly compared with healthy control( 32. 77 ± 4. 28% and 18. 53 ± 3. 27%,P 0. 01 and P 〈0. 001). Compared with control,the level of soluble CD28 was increased significantly in serum of type2 diabetes patients(( 2. 86 ± 0. 96) vs( 0. 61 ± 0. 19) μg /L,P 〈0. 001). Conclusion The costimulatory molecule CD28 maybe take part in the immunological pathogenesis of type 2 diabetes.
