BMSCs移植后脊髓损伤大鼠VEGF表达的变化

摘    要:目的观察骨髓间充质干细胞移植对大鼠脊髓损伤后血管内皮生长因子基因和蛋白表达的影响。方法以贴壁法分离培养大鼠骨髓间充质千细胞。压迫法制作大鼠脊髓压迫损伤模型。假手术组仅咬除T8-10棘突和椎板,不损伤脊髓。模型制作后DMEM组、骨髓间充质干细胞组分别进行DMEM与骨髓间充质干细胞损伤区周围局部4点注射。结果假手术组脊髓组织中仅见微量血管内皮生长因子表达。骨髓间充质干细胞组在细胞移植后1,3,5 d,血管内皮生长因子mRNA表达趋势与DMEM组相同,但表达水平均明显高于相应时间点的DMEM(P0.05),移植后7,14 d血管内皮生长因子表达仍明显高于DMEM组(P0.01)。结论骨髓间充质干细胞移植增加了脊髓损伤后血管内皮生长因子基因和蛋白的表达,并延长其表达时间。

Efects of bone marrow mesenchymal stem cells transplantation on vascular endothelial growth factor expression in rats after spinal cord injury

Abstract:Objective To observe the effects of BMSC transplantation on the expressions of vascular endothelial growth factor gene and protein in rats after spinal cord injury. Methods Rat BMSCs were isolated and cultured by adherence method. Rat models of spinal cord compression injury were induced by compression method. Spinons process and vertebral plate of thorax 8 - 10 spinal cord were removed, but did not injure the spinal cord in the sham operation group. Rats received separately four injections of DMEM or BMSCs in lesion area in the DMEM group and BMSCs group after spinal cord injury. Results VEGF expression was presented only in microamount endothelial cells of blood vessels in the sham operation group. The tendency of VEGF mRNA expression in the BMSCs group were the same as the DMEM group at days 1,3 and 5 after cell transplantation, but the levels of VEGF mRNA expression were significantly higher than in the DMEM group at coresponding time points ( P 〈 0.05). VEGF expression in the BMSCs group was significantly higher than in the DMEM group at days 7 and 14 after transplantation ( P 〈 0. 01 ) . Conclusion BMSC transplantation increases VEGF gene and protein expression and extends its expression time after spinal cord injury.
