锌对糖尿病大鼠视网膜VEGF表达的影响

摘    要:目的研究金属锌对大鼠糖尿病视网膜病变(diabetic retinopathy,DR)血管内皮细胞生长因子(VEGF)表达的影响,证实锌对糖尿病视网膜病变的防治作用。方法用Wistar大鼠建立糖尿病视网膜病变模型,随机分成4组:正常对照组、正常补锌组、高糖组、高糖+锌组。3M后轻摘眼球,在视网膜切片上行VEGF免疫组化。结果正常对照组、正常+锌组及高糖+锌组视网膜石蜡切片中免疫组化染色VEGF阳性表达,高糖组视网膜石蜡切片VEGF免疫组化明显阳性表达,高糖组视网膜切片VEGF免疫组化IOD值与正常组、正常+锌组及高糖+锌组相比明显增高(P0.001),正常组视网膜切片VEGF免疫组化IOD值与正常+锌组及高糖+锌组相比无显著性差异(P0.05)。结论我们认为补充金属锌能减少糖尿病大鼠视网膜病变VEGF的表达,有防治DR的作用。

Effect of Zinc on Vascular Endothelial Growth Factor expression in Retinopathy of diabetic rats

Abstract:Objective To research preventative and therapeutic effects of zinc on vascular endothelial growth factor(VEGF)in retinopathy of diabetic rat.Methods 40 Wistar rat were divided randomly into 4 groups,The normal control group、the normal with zinc group、the diabetic retinopathy rat model group、the diabetic retinopathy rat model with zinc group.After 3 month,eyeballs were picked,and immunohistochemistry staining for VEGF were done on retinal paraffin slide.Results Immunohistochemistry staining for VEGF showed positive expression in all group,The diabetic retinopathy rat model group is higher than other groups,which have significant difference(P0.001).The control group、the normal with zinc group and diabetic retinopathy rat model with zinc group do not have statistical difference(P0.05).Conclusion We think the supplemen of zinc have inhibition effect on VEGF expression,the supplemen of zinc have preventative and therapeutic effects on diabetic retina rat.
