摘    要:目的 探讨小鼠骨髓间充质干细胞经脉冲电磁场辐射不同时间后其细胞增殖水平的变化。方法 用密度梯度离心法分离小鼠骨髓间充质干细胞 ,再经贴壁筛选法筛选 ,对生长良好的第 3代小鼠骨髓间充质干细胞进行不同时间的脉冲电磁场辐射 (频率 50Hz、正弦波形、不同强度 ) ,用MTT法、流式细胞仪法测定细胞生长增殖水平及细胞周期变化。结果 小鼠骨髓间充质干细胞经脉冲电磁场辐射 3d(每天 2次 ,每次 3 0min)后 ,各实验组细胞增殖水平均有明显提高 ,差异有显著性意义 (P <0 .0 5)。采用流式细胞仪测定细胞周期 ,发现各实验组 (S G2 /M )期细胞数量较对照组均有不同程度的增长 ,差异有显著性意义 (P <0 .0 5)。各组均未发现DNA倍体异常细胞。结论 小鼠骨髓间充质干细胞经频率为 50Hz、正弦波形、强度分别为 4mT ,3mT ,2mT ,1mT的脉冲电磁场辐射 3d(每天 2次 ,每次 3 0min ,间隔 12h)后 ,能明显促进该细胞体外培养的增殖水平

Abstract:Objective To study the effect of pulsed el ectromagnetic fields (PEMFs) on the proliferation of bone mesenchymal stem cells in mice. Methods The bone marrow mesenchymal stem ce lls were obtained by using gradient centrifuge method, and then selected by the adhesive method. The third generation cells were irradiated by use of pulsed el ectromagnetic fields of 50Hz,sinusoid wave, at different intensities and for dif ferent time. The method of MTT was employed to evaluate the level of proliferati on. The parameters regarding the variation of the cell cycle were detected with the flow cytometry (FCM). Results The proliferation o f the bone mesenchymal stem cells in mice was greatly accelerated after 3 days o f irradiation by pulsed electromagnetic fields of 50Hz,sinusoid wave (twice dail y, 30 min each time ) at the intensities of 4mT,3mT,2mT and 1mT. The percentage of cells at the (S G2/M) phase was increased significantly ( P <0.05). No abno rmal ploidy was found in every group. Conclusion Thre e days of irradiation (twice daily, 30 min each time) of pulsed electromagnetic fields of 50Hz,sinusoid wave at the intensities of 4mT, 3mT, 2mT and 1mT could i ncrease the proliferation of bone mesenchymal stem cells of mice in vitro.
