碱性成纤维细胞生长因子对大鼠脊髓损伤后神经细胞凋亡的抑制

摘    要:目的探讨大鼠脊髓损伤后应用碱性成纤维细胞生长因子(basicfibrob-1astgrowthfactor,bFGF)对脊髓损伤区细胞凋亡的影响.方法利用Allen氏WD(Weightdrop,WD)技术,以10 g×2.5 cm致伤力造成SD大白鼠T8脊髓损伤模型,并于损伤平面以下蛛网膜下腔置细塑料导管.bFGF治疗组(A组)分别于术后即刻,1,2,4,8,12,24,及48h经导管注入bFGF溶液20μL(含bFGF100u),以后每周经导管注入20 μL bFGF;对照组(B组)则在同时间注入等量生理盐水.损伤后1,3,7,14,28d对脊髓损伤区进行细胞凋亡的检测(TUNEL),采用计算机图像分析技术进行定量分析并计算凋亡指数(AI),AI=凋亡细胞核数/总细胞核数计算.结果A,B两组中均发现凋亡细胞,A组损伤后不同时间段神经细胞凋亡指数分别为(4.57±0.43),(538±1.16),(3.43±0.65),(3.38±0.58),(2.63±0.43);B组分别为(7.36±0.68),(13.96±2.74),(9.26±1.03),(7.25±0.73),(5.79±0.57).B组细胞凋亡率大于A组结论bFGF能抑制脊髓损伤后脊髓损伤区的细胞凋亡.

Abstract:AIM: To explore the inhibition effect of basic fibroblast growth factor(bFGF) on apoptosis following spinal cord injury in rats.METHODS: SD rats were suffered (10 g ×2.5 cm)impaction to prepare a T8 spinal cord injury model on following the technique of Allen Weight drop (WD) method. A thin plastic tube was gently inserted into the subarachnoid space below the injury level. In the bFGF group(group A), 20 μL bFGF (bFGF 100 u)was injected shortly after the injury and 1,2,4,8,12,24 and 48 hours post the operation via the tube; bFGF was injected once per week at dosage of 20 μL; The saline group(group B): 30 spinal cord injury animal models were prepared, the same volume of normal saline was given to replace bFGF at the same period as control. TUNEL technique was used to determine apoptosis cells at 1, 3, 7, 14, 28 days after the injury. Apoptosis index(AI, AI = Number of apoptosis cell' s nucleus/Number of total cells' nucleus)was calculated by a computer image quantitative analysis system.RESULTS: Apoptosis cells were observed in both of the two groups. AI at different time period was (4.57±0.43), (5.38 ±1.16), (3.43±0.65), (3.38±0.58), (2.63±0.43) in group A; and (7.36 ±0.68), (13.96±2.74), (9.26±1.03), (7.25±0.73), (5.79±0.57)ingroupB. AI of group B was apparently higher than that of group A.CONCLUSION: bFGF can inhibit cell apoptosis in the injured area of spinal cord.
