摘    要:目的:TIPE 蛋白在肿瘤发生发展中起重要作用,但目前为止其在宫颈癌中的表达情况及潜在生物学功能还 未见报道,本研究旨在探讨TIPE 在宫颈癌中的表达水平及对宫颈癌细胞生物学功能的影响和可能机制。方法:免疫组化检 测宫颈癌组织及癌旁组织中TIPE 的表达,分析表达水平与患者预后的相关性;细胞实验检测TIPE 对Siha 细胞增殖影响; Western blot 实验初步探讨其可能的分子机制。结果:宫颈癌组织中TIPE 表达水平较癌旁组织明显上调,且与预后负相关;细 胞实验发现其促进宫颈癌细胞系Siha 的增殖;机制研究发现TIPE 抑制p53 乙酰化,从而抑制其下游分子p21 的表达。结论: TIPE 在宫颈癌组织中表达上调,且通过抑制p53 乙酰化促进宫颈癌细胞增殖,提示其参与了宫颈癌的发生发展进程。

TNFAIP8 ( TIPE ) promotes proliferation of cervical cancer by suppressing#br# p53 acetylization
Abstract:Objective:TIPE plays important roles in the development of carcinomas.However,the expression pattern and biological effect of TIPE in cervical cancer have not been elucidated.The aim of the study is to investigate the expression of TIPE in cervical cancer and to explore its biological effects and possible mechanisms.Methods: The expression of TIPE in cervical cancer or adjacent tissues was detected by immunohistochemistry.The correlation between the expression levels and the prognosis of the patients was analyzed as well.The effect of TIPE on the proliferation of Siha cells were detected using cell growth curve,clone forming and cell cycle methods.Western blot was used to determine the possible molecular mechanism.Results: The expression of TIPE in cervical cancer tissue was significantly up-regulated compared to control and negatively correlated with prognosis.Moreover,TIPE promoted the proliferation of cervical cancer cells.Mechanically,TIPE could inhibit the acetylation level of p53,by which suppresses the p21 expression.Conclusion: Up-regulated expression of TIPE in cervical cancer tissues promotes the proliferation of cervical cancer cells by inhibiting p53 acetylization,suggesting that TIPE is involved in the development and progression of cervical cancer.
