高压氧对小鼠脑缺血再灌注损伤的保护作用

摘    要:目的探讨高压氧(HBO)对脑缺血再灌注小鼠神经元的保护作用。方法选取昆明小鼠27只,采用无创微动脉夹阻断双侧颈总动脉血流的方法建立脑缺血再灌注模型。采用HE染色、透射电镜观察脑缺血再灌注小鼠神经元的损伤及HBO干预后的改变,结果光镜显示3组小鼠额叶皮层及海马有不同程度的神经细胞变性、坏死;实验组与对照组异常细胞数比较羞异有统计学意义(P〈0.05):同组小鼠的皮质和海马比较差异无统计学意义(P〉0.05)。电镜观察显示再灌注后额叶皮质及海马有不同程度的神经元变性、坏死,实验组损害明显减轻,与对照组比较差异有统计学意义(P〈0.05);同组小鼠的皮质和海马损伤程度比较差异无统计学意义(P〉0.05)。结论HBO治疗可对小鼠脑缺血再灌注损伤具有保护作用.可促进损伤的修复。

Abstract:Objective To study the protective effect of hyperbaric oxygen (HBO) on neuron of ischemia reperfusion brain injury in mice. Methods Twenty-seven Kunming mice were randomly divided into 3 groups:hyperbaric oxygen group,control group and sham operation group. Cerebral ischemia reperfusion models were established by clamping both carotid arteries in mice. The 0.20 MPa HBO were applied 10 times after operations on hyperbaric oxygen group. Using HE stains and transmission electron microscope,histopathological changes of nerve cell structure in cortex and hippocamal tissue were observed. Results With light microscope the varying degree of neural denaturation and necrosis in cortex and Hippocampus issue were found after cerebral ischemia reperfusion. In the control group,necrosis of neurons was obvious,the number of abnormal cells in control group was more than that in hyperbaric group,the difference was significant (P 〈 0.05). The varying degree of neural denaturation and necrosis 'after ischemia reperfusion was found by electron microscope. Compared with the control group,the neurous were injured slightly in hyperbaric oxygen group with significant difference (P 〈 0.05).While in the same group,injuried cells in cortex and hippocampus issue,the difference was not significant (P 〉 O.05).Conclusion It is demonstrated that HBO could reduce the intensity of brain injury after cerebral ischemia reperfusion and it could improve the repair activity and restore the function of neurons. Thus the HBO may play an important role in repairing and protective function for central nervous system.
