用荧光原位杂交方法检测肺癌印片标本中性染色体数目的异常

目的 研究肺癌细胞中性染色体拷贝数的异常。方法 取新鲜的肺癌手术标本作印片，利用双色荧光原位杂交（ＦＩＳＨ）技术对１６例肺癌印片标本（其中男性病例１２例，女性病例４例）中Ｘ染色体和Ｙ染色体数目进行检测。结果 发现１２／１６（７５％）的标本中有Ｘ染色体数目增多，其中女性患者３例，男性患者９例；在１２例男性患者中，有１例发生Ｙ染色体数目增多，有５例（４１．７％）发生Ｙ染色体数目减少。总计出现性染色体数

SEX CHROMOSOME COPY NUMBER ABERRATIONS IN LUNG CANCER TOUCH PREPARATIONS DETECTED BY FISH

Objective\ The aim in this paper was to prepare anti P 53 monoclonal antibody, which can specifically bind to human P 53 , and made it widely useful in P 53 detection by immunohistochemistry(IHC). Methods\ The P 53 DNA fragment encoding N terminal 180 amino acid was obtained by PCR and was cloned into P G EX 2T plasmid expressing glutathione S transferase(GST); The P 53 GST fusion proteins expressed by JM109 was used for immunizing BALB/c mice. Results\ We have raised one hybridoma strain secreting McAb to human P 53 (named M126); the IHC analysis of 52 praffin embedded sections from human breast cancer with M126 and P AB 1801 has shown that the positive immunoreactions were shown in 25 cases (48 1%) and 22 cases (42 3%) respectively; the staining of M126 was stronger and preferable to import P 53 McAb P AB 1801. Conclusion\ M126 can be used instead of P AB 1801 for studying immunohistochemical analysis on P 53 protein.
