超剂量米非司酮对大鼠子宫内膜超微结构的影响

摘    要:目的 :电子显微镜下观察超剂量使用米非司酮的大鼠子宫内膜超微结构改变 ,为进一步扩大米非司酮临床使用范围提供形态学依据。方法 :实验组大鼠每日灌服米非司酮 30 m g/只 ,分别于第 8,15 ,2 9天处死取子宫 ,超薄切片 ,电子显微镜观察。结果 :子宫内膜纤毛细胞纤毛减少 ,分泌细胞胞质内出现有空泡的圆形颗粒 ,并有顶浆分泌 ,内膜上皮基质呈动情前表现。子宫腺细胞基底及核上区有糖原斑 ,为动情晚期形态。小血管内皮间隙增大 ,其周围大量多形核细胞浸润 ,似人类月经期崩解的子宫内膜。子宫内膜多种细胞及间质内均可见多泡体、巨大溶酶体和残余体。结论 :长期超剂量使用米非司酮会使子宫内膜发育不同步 ,还可能导致细胞代谢失调

Abstract:Objective: To provide trustworthy morphological evidence for widening the range of using mifepristone clinically the ultrastructural changes inducedby overdosage of mifepristone were observed under electron microscope. Methods:30 mg mifepristone was given to each experimental rats intragastrically the rats were killed on surviving 8, 15, 29 days separately, then endometrium was taken out and made into sections for observing under the electron microscope. Result: The number of the cilia of the ciliated cells reduced obviously the vacuolization lysosome presented in cytoplasm of the serectory cells, the apocrine protruded from the top of the cells, this shown that the endometrium was in the early proliferation phase. Glycogen densa in the cells of the uterine gland appeared at the base or at the area above the nucleus, this features indicated that these cells stayed in early secretory stage. The gap between epithelial cells of the small vessels became widened and a large number of infiltrated lcukocytes around the vessels were found like the disintegrated endometrium in menstrual phase. Multivisicular body and large residual body existed in endometrium cells and in various intercellular substance. Conclusion: Taking overdosage of mifepristone for a long period of time would making the endometrium to develop asynchrinously and to mctabolize disorderly.
