MACC1基因在脑胶质瘤中的表达及其对U87细胞凋亡和增殖的影响

摘    要:目的探讨基因与脑胶质瘤的相关性及其表达改变对胶质瘤U87细胞凋亡和增殖能力的影MACC1响。方法荧光定量PCR检测45例脑胶质瘤及相应癌旁组织中MACC1基因的表达;设计并合成基因特异MACC1性的siRNA,转染U87细胞;Western blot检测转染后MACC1蛋白的表达;双标流式细胞法检测细胞凋亡;MTT法测定细胞生长抑制率。结果基因在脑胶质瘤组织中表达明显上调;转染基因特异性的siRNA后MACC1蛋白表达明显降低,凋亡明显增加,生长抑制率明显增加,细胞增殖活力明显降低。结论 MACC1基因的表达异常与脑胶质瘤的发生相关,抑制其表达能促进肿瘤细胞的凋亡同时抑制细胞增殖。

Abstract:Objective To research the relationship of the expression changes of metastasis-associated in colon cancer-1(MACC1) gene with brain glioma, and the influence of its expression to apoptosis and proliferation in U87 cell.
