龟板对脊髓损伤大鼠神经干细胞的作用

摘    要:为了观察龟板对大鼠脊髓损伤后 nestin表达和后肢功能恢复的影响 ,本实验于手术后 1d、7d、14 d、2 1d、2 8d分别对大鼠进行 Tarlov评分和斜板试验检查后肢功能 ,处死动物后应用免疫组织化学技术检测神经上皮干细胞蛋白的表达 ,用图像分析仪进行定量分析 ,观察了龟板对脊髓损伤后神经上皮干细胞蛋白的影响。结果表明 ,脊髓损伤后第 1d,在两组受损伤脊髓灰质中都可见到 nestin的表达 ;第 14 d时达到高峰 ,龟板组为 3 8.2 4± 6.83 ,损伤组为 2 2 .48± 7.83 (P<0 .0 5 )。龟板组可使损伤的脊髓中 nestin持续高表达至术后第 2 8d,而损伤组仅持续高表达至术后 2 1d。增加的 nestin阳性细胞数量与神经功能的改善平行。本研究结果提示 :龟板可减轻神经损伤症状和促进脊髓损伤后神经干细胞的增殖

Abstract:The aim of the present study is to investigate the effect of Tortoises shell on neural stem cell following injured spinal cord as well as functional recovery in the rats. After operation open field and inclined plane tests were performed to test the function of the rear limbs, the animals were sacrificed 1, 7, 14, 21 and 28 day after surgery. By detecting expression of the neuroepithelial stem cell protein, nestin using immunohistochemical method, the effects of Tortoises shell on neural stem cell following injured spinal cord were observed. The nestin positive cells were detected in the gray matter one day after spinal cord injury(SCI), and increased after operation and reached their peak at day 14 after SCI in both group A and B. The number of the nestin positive cells was 22.48±7.83 and 38.24±6.83(P<0.05). The positive cells in B group maintained at high level to day 28; however, in group A the positive cells decreased significantly at day 28. The group B had a significant higher value than that in group A. The increase in the nestin positive cells was paralleled by a significant improvement in neurological function evaluated by open field and inclined plane tests. These results suggest that Tortoises shell can alleviate the neurological symptoms and promote the proliferation of neural stem cell following spinal cord injury of rats.
