高压、高压氧对小鼠大脑酶活性的影响——组织化学研究

摘    要:本文研究高压和高压氧对小鼠大脑碱性磷酸酶等6种酶的活性影响。30只小鼠随机分为空白对照组、高压组和高压氧组。高压组动物置于加压舱内,高压氧组动物置于同一加压舱中的小套舱内,分别给予5ATA的空气和纯氧,停留37分钟。高压处理结束,按组织化学要求取材、固定、切片和酶活性反应。实验结果表明:高压氧可致动物小痉挛;高压环境对动物机体无明显刺激影响;高压氧使碱性磷酸酶(AKP)、葡萄糖-6-磷酸酶(G-6-P)、乳酸脱氢酶(LDH)、乙酰胆碱酯酶(AChE)活性增强,高压环境使上述4种酶活性略有增强,和空白对照组比差别不明显。高压和高压氧使单胺氧化酶(MAO)和过氧化物酶活性略有降低。本文对高压氧环境导致机体行为变化和6种酶活性改变的意义作了讨论。

Abstract:We have studied mainly the effects of hyperbaric air and oxygen on the activity of AKP and other five enzymes in mouse brain with histochemical methods. 30 mice were randomly divided into three groups: the blank control group; the hyperbaric air experimental group; and the hyperbaric oxygen experimental group. The animals of the two experimental groups were placed in the same hyperbaric animal chamber, in which the animals of hyperbaric oxygen group were put in a small case separa tely. These animals were given air and 100% oxygen at 5ATA respectively. The animals of the two experimental groups were kept at 5ATA for 37 minutes. As soon as high pressure was reduced to normal pressure, cerebral tissues were removed and fixed according to the method for histochemisry. Then samples were sectioned and processed for the reaction activity of enzymes. The experimental results showed that in animals under hyperbaric oxygen environment small convulsion was observed but no effects were seen on the experimental animals under air condition at 5ATA. Hyperbaric oxygen increased the reaction activity of alkaline phosphatase(AKP), glucose-6-phosphatase (G-6-P), lactate dehydrogenase (LDH), and acetylcholinesterase(AChE) of cerebral tissue significantly. Hyperbaric air increased reaction activity of the above four enzymes of brain tissues by a little. Hyperbaric air and hyperbaric oxygen reduced reaction activity of monoamine oxidase (MAO) andperoxidase of cerebral tissues by a little.The significance of behaviour changes of the experimental animals and reactivity change of six enzymes of brain tissue under hyperbaric oxygen condition were discussed
