摘    要:目的:探讨沉默RWDD3表达对人胶质瘤U251细胞生物学特征的影响。方法:构建靶向RWDD3的shRNA重组慢病毒并转染U251细胞,通过real-time PCR和Western blot在mRNA及蛋白水平鉴定转染结果。MTT法检测转染后细胞的细胞活力;平板克隆实验检测克隆形成能力;Brd U实验检测细胞增殖能力;Transwell实验检测细胞迁移和侵袭能力的变化;流式细胞术检测细胞周期及细胞凋亡的改变。结果:real-time PCR和Western blot结果均表明成功建立稳定沉默RWDD3的U251细胞株。与空白对照组及阴性对照组比较,转染RWDD3-shRNA组的细胞活力和克隆形成能力降低;侵袭及迁移能力均下降,穿膜细胞数明显减少;细胞周期进展被抑制,阻滞于G0/G1期,细胞凋亡率增高(P0.05)。结论:RWDD3在胶质瘤细胞增殖和侵袭过程中发挥重要作用,提示RWDD3有望成为人脑胶质瘤基因治疗的候选靶点。

Abstract:AIM: To investigate the effect of RWDD3 gene silencing on the biological characteristics of human glioma U251 cells.METHODS: A lentiviral vector expressing RWDD3 shRNA was constructed and transfeeted into the U251 cells. The expression of RWDD3 at mRNA and protein levels was detected by real-time PCR and Western blot, respectively. The cell activity was determined by MTT assay. The colony formation ability was detected by the colony formation assay. The cell proliferation ability was detected by BrdU incorporation assay. The cell invasion and migration were evaluated by Transwell assay. Flow cytometry was used to monitor the changes of cell cycle distribution and apoptosis.RESULTS: Recombinant lentivirus was successfully transfected into U251 cells. Compared with the cells transfected with the scrambled shRNA and control cells, the cell activity, colony formation ability, and the invasive and migratory activities were inhibited, the cell cycle was arrested in G0/G1 phase, and the apoptosis was increased in the U251 cells transfected with RWDD3 shRNA(P<0.05).CONCLUSION: RWDD3 plays a vital role in proliferation and invasion of glioma cells. It may serve as a potential target of gene therapy for glioma.
