内皮素-1在大鼠肝脏冷保存不同时相的含量变化及其与肝损伤的关系

摘    要:目的:研究内皮素- 1 在大鼠肝脏冷保存不同时相的含量变化及其与门静脉灌注压及肝损伤的关系。方法:以离体鼠肝灌流为模型,用放免方法检测肝脏保存不同时间灌流液及肝脏中内皮素- 1(ET- 1) 的变化,同时观察门静脉灌注压及肝脏功能变化,并做相关分析。结果:随保存时间延长,ET- 1 水平呈进行性升高。ET- 1 与门静脉灌注压变化呈明显正相关( r = 0-970 ,P< 0-01) ,与胆汁分泌量呈明显负相关( r = - 0-864 ,P< 0-05) ,与灌注液AST、ALT、LDH 也呈明显正相关( r= 0-919 ,P< 0-01) 。结论:ET- 1 参与并加重肝脏保存再灌注损伤,尤其对肝脏微循环的直接影响。因此ET- 1 很可能是形成该损伤的重要内源性致伤因子之一。

Abstract:AIM:To study the changes of endothelin-1 levels in cold ischemia/reperfusion injury of rat liver and its relation to portal perfusion pressure and hepatic functions.METHODS:Using radioimmunoassay, we measured endothelin-1 levels in effluent and liver during different periods of cold preservation in isolated perfused rat liver model. In the meantime, we analysed the correlation of endothelin-1 changes with portal perfusion pressure and hepatic functions. RESULTS:Endothelin-1 levels enhanced markedly as the prolonged time of preservation. It was quite apparent that endothelin-1 levels showed positive correlation to portal perfusion pressure( r=0.970, P<0.01 ) and aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase contents in effluent ( r=0.919,P<0.01 ), negative correlation to bile flow ( r=-0.864, P<0.05 ). CONCLUSION:Endothelin-1, as one of the important endogenous traumatogenic factors, participate in and aggravate hepatic injury during preservation reperfusion, especially impact the hepatic microcirculation directly.
