摘    要:目的通过观察胎盘组织匀浆上清(PTHS)对小鼠脾脏树突状细胞(DC)生物学特征的影响。探讨妊娠免疫耐受的机制。方法分离妊娠及正常BALB/C小鼠脾脏单个核细胞,诱导分化为树突状细胞,经PTHS作用后,流式细胞仪检测树突状细胞的表面标志和抗原摄取能力,MTr法检测其同种异基因淋巴细胞刺激能力。ELISA方法检测其产生的细胞因子水平。结果妊娠小鼠脾脏树突状细胞的生物学特征与正常对照小鼠比较差异无统计学意义,而经PTHS作用后的妊娠小鼠及正常对照小鼠的脾脏CDIIb+树突状细胞明显降低。而CD8α+树突状细胞明显增高(P〈0.01);树突状细胞的抗原摄取能力及刺激同种异基因的淋巴细胞反应的能力也明显低于正常小鼠(P〈0.01);PTHS作用后的树突状细胞产生较高水平的IL-10和较低水平的IL-12,与对照组比较差异有统计学意义(P〈0.01)。结论PTHS可向下调节BALB/c小鼠脾脏树突状细胞的生物学特征,可能与妊娠免疫抑制有关。

Influence of placental tissue extract on the growth of splenic dendritic cells of BALB/c mice

Abstract:Objective To study the biological characteristics of dendritic cells grown in medium with placental tissue extract. Methods Splenic mononuclear cells of BALB/c mice were isolated and then induced to differentiate to dendritic cells. After treatment with the placenta tissue extract, the phenotype and the capability of antigen uptake by the dendritic cells were determined with flow cytometry. Capability of stimulating allogeneic lymphocyte proliferation was detected by the methyl thiazoly tetrazolium (MTY) method. The level of cytokines was measured by enzyme linked immunosorbent assay. Results The biological characteristics of dendritic cells prepared from both the normal and pregnant mice were similar (P〉0.05). After treatment with the placental tissue extract, dendritic cells from both the normal and pregnant mice had a similar pattern of lower percentage of CDllb+ cell and an increased percentage of CD8ct+ cells, a lower capability of antigen uptake and stimulation of allogeneic lymphocyte proliferation, and secreting more IL-10 and less IL-12, when compared with the normal untreated control mice (P〈0.01). Conclusion The reduced expression of the biological characteristics of splenic dendritic ceils in BALB/c mice may be related to the immunosppressive functions of the tissue extract.
