睫状神经营养因子对视神经损伤修复作用的研究

摘    要:目的:观察睫状神经营养因子(CNTF)能否促进视神经(ON)损伤后在原中枢神经环境中再生,为临床救治提供理论依据和新的治疗途径。方法:选用成年日本青紫蓝兔22只,其中随机抽取两只做正常对照,不做任何处理;其余20只为实验组,构造视神经中度损伤模型(中号显微血管夹于视神经后3 mm处夹持20 s)。于损伤后即时、3 d、7 d、14 d各时间点给予右眼(实验眼)玻璃体腔内注射CNTF 2μl(1μg/μl),左眼(对照眼)于同等时间点注射等量双蒸水。于损伤后3 d、7 d、14 d、28 d分别取实验眼和对照眼视神经行常规HE染色观察视神经组织学变化;免疫荧光检测观察生长相关蛋白-43(GAP-43)在视神经的表达;同时将动物实验眼及对照眼于损伤前、损伤后即刻及处死前进行视觉诱发电位(F-VEP)检查以观察兔视功能变化情况,采用SPSS 13.0统计学软件包对数据进行统计学分析。结果:HE及免疫荧光检测实验组组织学变化明显好于对照组;F-VEP检查可见视神经损伤后即时闪光视觉诱发电位波形几近熄灭。伤后1周,两组潜伏期基本恢复至损伤前水平,振幅恢复缓慢,组间比较差异无统计学意义(P〉0.05);2周以后两组间比较差异有统计学意义(P〈0.01)。结论:睫状神经营养因子对视神经损伤有修复作用,可促进视神经轴突在原中枢环境中的再生。
