壳聚糖对镉染毒大鼠肾脏中超氧化物歧化酶活性和丙二醛含量的影响

摘    要:
目的探讨壳聚糖对镉染毒大鼠肾脏中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。方法50只SD大鼠,雌雄各半,随机分成5组,分别为对照组、镉染毒组(1.0 mg.kg-1CdCl2)、壳聚糖低剂量干预组(20 mg.kg-1壳聚糖+1.0 mg.kg-1CdCl2)、壳聚糖中剂量干预组(100 mg.kg-1壳聚糖+1.0 mg.kg-1CdCl2)和壳聚糖高剂量干预组(500 mg.kg-1壳聚糖+1.0 mg.kg-1CdCl2)。对照组先用生理盐水灌胃,2 h后腹腔注射生理盐水;镉染毒组先用生理盐水灌胃,2 h后腹腔注射CdCl21.0 mg.kg-1;各壳聚糖干预组分别采用20、100、500 mg.kg-1壳聚糖灌胃,2 h后再分别腹腔注射CdCl21.0 mg.kg-1,每日1次,连续7 d。7 d后取肾脏测定肾脏脏器系数、SOD活性及MDA含量。结果镉染毒组肾脏脏器系数及MDA含量大于对照组,SOD活性则小于对照组,差异均有统计学意义(P<0.05)。壳聚糖高剂量干预组肾脏脏器系数低于镉染毒组(P<0.05);壳聚糖各剂量干预组的肾脏SOD活性高于镉染毒组,MDA含量低于镉染毒组,差异均有统计学意义(P<0.05)。结论在本实验条件下,1.0 mg.kg-1剂量的CdCl2对SD雄性大鼠具有肾脏毒性;大于20 mg.kg-1剂量的壳聚糖对1.0 mg.kg-1剂量的CdCl2所致SD大鼠肾脏的氧化损伤有拮抗作用。

Abstract:
Objective To investigate the effect of chitosan on superoxide dismutase(SOD) activity and malondialdehyde(MDA) content in kidney of rat exposed to cadmium. Methods Fifty cases of SD rats were divided into five groups randomly,control group,cadmium infected group(1.0 mg·kg-1 CdCl2),chitosan low dose intervention group(20 mg·kg-1 chitosan +1.0 mg·kg-1 CdCl2),chitosan middle dose intervention group(100 mg·kg-1 chitosan +1.0 mg·kg-1 CdCl2) and chitosan high dose intervention group(500 mg·kg-1 chitosan +1.0 mg·kg-1 CdCl2) respectively.Rat stomachs were perfused with normal saline(NS),and intraperitoneal injection of NS after 2 hours in control group.NS were filled into stomachs first and intraperitoneal injection of 1.0 mg·kg-1 CdCl2 after 2 hours in cadmium infected group.20,100 and 500 mg·kg-1 of chitosan were filled into stomachs,and intraperitoneal injection of 1.0 mg·kg-1 CdCl2 after 2 hours in each chitosan intervention group,one times a day for 7 days.The organ coefficient,SOD activity and MDA content in kidney were determined after 7 days. Results The organ coefficient and MDA content in cadmium infected group were higher than those in control group,but SOD activity was lower than that in control group(P<0.05).The organ coefficient of kidney in chitosan high dose intervention group was lower than that in cadmium infected group(P<0.05).The SOD activity of kidney in each chitosan intervention group were higher than that in cadmium infected group,and the MDA contents were lower than that in cadmium infected group(P<0.05). Conclusion Under this experimental conditions,1.0 mg·kg-1 CdCl2 can be induce kidney toxicity in rats,and when the dose of chitosan more than 20 mg·kg-1,chitosan have antagonistic effects to kidney oxidative damage in rats induced by 1.0 mg·kg-1 dose of cadmium.
