不同浓度葡萄糖对大鼠大脑皮层神经元凋亡的影响

摘    要:目的探讨不同浓度葡萄糖对大鼠大脑皮层神经元凋亡的影响。方法取新生1 d内SD大鼠的大脑皮层,皮层神经元经原代培养,通过胰蛋白酶消化法获取单细胞悬液,免疫荧光染色鉴定神经元纯度,然后随机分为正常组(葡萄糖浓度为25 mmol.L-1)及50、75、100 mmol.L-1葡萄糖组,Hoechst染色检测细胞凋亡,噻唑蓝比色法检测各组神经元活性。结果与正常组和50 mmol.L-1葡萄糖组相比,75、100 mmol.L-1葡萄糖组大鼠大脑皮层神经元凋亡率明显升高(P<0.01),活性显著下降(P<0.01);且100 mmol.L-1葡萄糖组大鼠大脑皮层神经元凋亡率高于75 mmol.L-1葡萄糖组(P<0.01),大鼠大脑皮层神经元活性低于75 mmol.L-1葡萄糖组(P<0.01)。结论高浓度的葡萄糖可引起大鼠大脑皮层神经元的凋亡,且神经元的凋亡情况呈剂量依赖关系。

Abstract:Objective To discuss the effect of glucose with different concentrations on apoptosis of rat cortical neuron. Methods The cortex of rat with one day old was obtained to primary culture the cortical neuron,the single cell suspension was received by enzyme digestion,the purity of neuron was identified by immunofluorescence,then the neurons were divided into normal group and glucose groups(50,75,100 mmol·L-1).The apoptosis of neuron was detected by Hoechst staining,and the activity of neuron was examined by methyl thiazolyl tetrazolium method. Results Compared with normal group and 50 mmol·L-1 glucose group,the apoptotic number of neuron increased and the activity of neuron decreased in the 75 mmol·L-1 glucose group and 100 mmol·L-1 glucose group(P<0.01).Compared with 75 mmol·L-1 glucose group,the apoptotic number of neuron increased and the activity of neuron decreased in the 100 mmol·L-1 glucose group(P<0.01). Conclusion The glucose with high concentrations can induce the apoptosis of cortical neuron of rat with dose-dependent manner.
