扶正解毒颗粒对染镍大鼠肾损伤的影响

摘    要:目的观察扶正解毒颗粒对硫酸镍染毒大鼠肾损伤的干预作用及其促排镍作用。方法将Wistar大鼠随机分为正常组、模型组、二巯基丁二酸(DMSA)组及扶正解毒颗粒大、中、小剂量组。除正常组外,其余大鼠以硫酸镍2.5 mg/(kg.d)腹腔注射染毒造模,扶正解毒颗粒大、中、小剂量组分别以扶正解毒颗粒5、10、20 mg/(kg.d)灌胃处理,DMSA组以DMSA 50 mg/(kg.d)灌胃,正常组与模型组予生理盐水灌胃。实验第4周末检测大鼠肾功能、血镍、尿镍水平,并进行肾脏组织病理学检查。结果随着造模时间的延长,扶正解毒颗粒各组大鼠血尿素氮、肌酐及血镍水平较模型组明显降低(P〈0.05和P〈0.01),尿中镍排泄增加(P〈0.05和P〈0.01),肾小管上皮变性、肾小管扩张、间质炎性细胞浸润等病理改变亦较模型组明显减轻。结论扶正解毒颗粒对染镍大鼠具有良好的促排镍作用,对镍暴露肾损伤有一定的拮抗作用。

Abstract:Objectives To study the interventing and nickel-excreting promoting effects of Fuzheng Jiedu granula on rats' renal damage induced by nickel.Methods Wistar rats were divided into six groups:normal group,model group,DMSA group,and low-,medium-,high-dose of Fuzheng Jiedu granula group.Model rats were induced with intraperitoneal injection of NiSO4 2.5 mg/(kg·d).Fuzheng Jiedu granula group was administered by gavage of Fuzheng Jiedu granula 5,10,20 mg/(kg·d).DMSA group was given DMSA 50 mg/(kg·d),and normal goup and model group were given normal saline.At the end of four weeks,the levels of BUN,SCr,blood nickel and urine nickel were measured and the renal pathological changes were observed,respetively.Results In Fuzheng Jiedu granula groups,BUN,Cr and blood nickel were all decreased(P〈0.05,P〈0.01),urine nickel increased obviously(P〈0.05,P〈0.01),and some pathological changes,such as the degeneration of renal tubularepithelial cell,the dilatation of tubule and interstitial infiltration of inflammatory cell appeared late and were attenuated as time progressed,compared with that of model group.Conclusion Fuzheng Jiedu granula can promote nickel excretion in rats exposed to nickel and has antagonistic effect on renal damage induced by nickel.
