摘    要:
目的 研究腹腔注射1—甲基—4—苯基—1,2,3,6—四氢吡啶(MPTP)对褐鼠黑质神经母细胞瘤凋亡相关的RNA结合蛋白(NAPOR)基因表达的影响,为阐明帕金森病(PD)的分子机制提供线索。方法 通过腹腔注射MPTP建立褐鼠PD模型,采用逆转录PCR方法研究NAPOR在黑质的表达情况。结果 通过MPTP腹腔注射后褐鼠模型出现典型PD症状、黑质酪氨酸羟化酶表达水平明显降低验证PD模型成功,PD鼠黑质NAPOR基因表达明显上调。结论 NAPOR为RNA结合蛋白,其表达的模式与神经元凋亡密切相关。提示MPTP对黑质神经元的毒性可能通过改变NAPOR基因的表达,促进神经元凋亡而实现。

Abstract:
Objective To study the effects on gene expression of NAPOR in the substantia nigra of C 57 BL mice and the molecule mechanism by peritoneal administration of MPTP. Methods The C 57 BL mice model of Parkinson's disease was prepared by peritoneal administration of MPTP. RT PCR technique was used to detect the different expression of NAPOR in the substantia nigra. Results The model was verified successfully by the typical PD symptoms and obviously reduced expression of gene tyrosine hydroxylase in substantia nigra after peritoneal administration of MPTP. The result of RT PCR for NAPOR revealed that there was higher expression in the substantia nigra of PD mice. Conclusion NAPOR is RNA binding protein. Its expression pattern is spatially and temporally coincident with neuronal apoptosis. The results suggest that toxicity of MPTP to neurons of substantia nigra may result from alteration of NAPOR expression and further promote apoptosis of neuron.
