| 摘 要： | 目的用pH梯度法制备重酒石酸长春瑞宾脂质体,考察不同因素对脂质体冻干前后包封率的影响。方法采用pH梯度法制备重酒石酸长春瑞宾脂质体,冷冻干燥,用Sephadex G-50柱层析法测定包封率,比较冻干前后包封率的大小。结果在药脂比为1∶40、孵育温度37℃、孵育时间30 min、洗脱液PBS(pH7.4,137 mmol/L NaCl,2.6 mmol/L KCl,6.4 mmol/L Na2HPO4,1.4 mmol/L KH2PO4)时,重酒石酸长春瑞宾脂质体冻干前后的包封率最高。结论药物与磷脂的比值、孵育温度和不同缓冲盐作为洗脱液,在制备过程中对重酒石酸长春瑞宾脂质体冻干前后的包封率都有不同程度的影响。 |
