健脾补肾药对脾虚大鼠环核苷酸及甲状腺激素水平的影响

摘    要:目的 :观察健脾补肾方药对实验性脾虚证大鼠环核苷酸及甲状腺激素水平的影响 ,探讨该方防治脾虚证的作用机理及脾与肾相互关系的意义。方法 :选用SD雄性大鼠 ,随机分为 4组 :即正常对照组 ,脾虚模型组 ,健脾补肾方高、低剂量组 ,采用大黄复制脾虚证动物模型 ,观察各组动物血浆环磷酸腺苷 (cAMP)、环磷酸鸟苷 (cGMP)及血清甲状腺素 (T4)、三碘甲腺原氨酸 (T3 )、促甲状腺素 (TSH)水平的变化。结果 :脾虚模型组大鼠血浆cAMP含量及cAMP/cGMP比值均比正常对照组升高 (p <0 0 1) ,T3 、T4含量比正常对照组下降 (p<0 0 1)。而健脾补肾方药能降低实验性脾虚证大鼠体内cAMP含量及cAMP/cGMP比值 (p <0 0 1) ,提高T3 、T4含量 (p <0 0 1)及脾脏、胸腺比值 ,又可使肾脏组织重量减轻等作用。 结论 :健脾补肾中药可以消除大黄的药物影响 ,防止出现脾虚症候 ,其作用机理与调节cAMP、cGMP及甲状腺激素水平有关。脾肾相关理论对临床有指导意义

Abstract:Objective To investigate the effect of JPBS recipe on the serum levels of cAMP, cGMP and thyroid hormones in PiXu rat models and to explore the treatment mechanism as well as the interrelationship between spleen and kidney. Methods 46 male SD rats were divided into 4 groups: controls (gavage with normal saline only, n=10), PiXu models (gavage with rhubarb decoction for 16 days, n=12), PiXu rats treated with high dose of recipe (gavage with recipe for the last 8 days, n=12), PiXu rats treated with low dose of recipe (gavage with half strength of recipe, n=12). Serum levels of cAMP, cGMP, T 3, T 4, TSH were determined in all the rats with RIA at the end of the experiment. Results Serum cAMP, cAMP/cGMP were significantly higher and serum T 3, T 4, TSH significantly lower in the PiXu rat models than those in the control gorup ( p <0 01). Treatment with the JPBS recipe could decrease the serum cAMP, cAMP/cGMP and increase the serum thyroid hormones significantly ( p <0 01 compared with PiXu models without treatment). Spleen and thymus body weight ratios were increased and kidney body weight ratio decreased. Conclusion JPBS recipe could antagonize the effect of rhubarb and ameliorate the PiXu symptoms in experimental rat models through the regalation of serum cAMP, cGMP and thyroid hormones levels. The interrelationship between spleen and kidney could be observed in the reciprocal changes of their body weight ratios.
