颌骨骨肉瘤中BMP-2mRNA的表达及其与淋巴结转移的关系

目的：观察颌骨骨肉瘤(osteosarcoma of the jaw，OSJ)中BMP-2 mRNA的表达，并探讨其与淋巴结转移的关系。方法：采用原位杂交方法．对21例OSJ标本进行BMP-2 mRNA检测。应用卡方检验．对OSJ中BMP-2 mRNA表达与淋巴结转移问的关系作统计分析。结果：21例OSJ中，17例BMP-2mRNA表达阳性(80.95％)，包括全部15例成骨细胞型和2例(2／4)成软骨细胞型OSJ。梭形或多边形的肿瘤细胞．细胞质中可见明显的阳性染色。12例出现淋巴结转移的OSJ标本，BMP-2mRNA表达均为阳性，而无BMP-2mRNA表达的OSJ标本中，无1例出现淋巴结转移。淋巴结阳性组，BMP-2mRNA的阳性表达率为100％；淋巴结阴性组，BMP-2mRNA的阳性表达率为55．56％．两者差异有统计学意义(P=0．021)。结论：BMP-2mRNA主要在成骨细胞型及部分成软骨细胞型OSJ中表达，其表达可能与OSJ的淋巴结转移存在一定关系。

The expression of BMP-2mRNA in osteosarcoma of the jaw and its relationship to lymph node metastasis

PURPOSE: To investigate the expression of BMP-2mRNA in osteosarcoma of the jaw(OSJ) and its correlation with the lymph node metastasis. METHODS:BMP-2mRNA expression in 21 cases of OSJ was detected by in situ hybridization. The correlation between lymph node metastasis and expression of BMP-2mRNA in OSJ was analyzed by Chi square test. RESULTS: The positive rate of BMP-2mRNA expression in 21 cases was 80.95% (17/21), including all osteoblastic (15/15) and 2 chondroblastic (2/4) OSJ. The positive staining was mainly localized in cytoplasm of fusiform and polygon tumor cells. All 12 cases with lymph node metastasis had positive staining, while those that displayed negative staining didn't have lymph node metastasis. The positive rate of BMP-2mRNA expression in patients with lymph node metastasis was 100%, while the positive rate in patients without lymph node metastasis was 55.56% .The difference between the two groups was significant statistically(P=0.021). CONCLUSIONS: The expression of BMP-2mRNA mainly occurred in osteoblastic and partly in chondroblastic OSJ. It may be related to lymph node metastasis of OSJ in this limited series.
