Wnt-1诱导分泌蛋白2对胃癌细胞上皮间质转化的影响*

摘    要:
的 探讨Wnt-1诱导分泌蛋白2(WISP2)在胃癌组织中的表达及其对胃癌患者癌细胞上皮间质转化的影响。方法 选取2016年3月—2018年5月四川绵阳四〇四医院收治的胃癌患者癌组织和癌旁组织标本,每组85例。采用免疫组织化学法检测WISP2在胃癌组织和癌旁组织中的表达情况;构建WISP2-shRNA和NC-shRNA稳定细胞系,采用细胞划痕实验检测WISP2下调对胃癌细胞迁移的影响;采用Transwell实验检测WISP2下调对胃癌细胞侵袭的影响;采用Western blotting检测WISP2下调对胃癌细胞E-钙黏蛋白(E-cadherin)、神经-钙黏蛋白(N-cadherin)和波形蛋白(Vimentin)表达的影响。结果 WISP2在胃癌组织(61.2%)中的阳性表达率高于癌旁组织(11.8%)(P?<0.05);与NC-shRNA组(84.74±9.61)%比较,WISP2-shRNA组(57.18±6.05)%细胞划痕愈合程度下降(P?<0.05);WISP2-shRNA组细胞侵袭个数(195.36±23.45)较NC-shRNA组(404.91±38.16)减少(P?<0.05);与NC-shRNA组比较,WISP2-shRNA组细胞E-Cadherin的表达水平升高(P?<0.05),N-Cadherin和Vimentin的表达水平下降(P?<0.05)。结论 WISP2在胃癌组织中高表达,下调WISP2的表达可抑制胃癌细胞的上皮间质转化和侵袭转移。

Abstract:
Objective To investigate the expression of Wnt-inducible secreted protein 2 (WISP2) in gastric cancer and its effect on epithelial-mesenchymal transition. Methods A total of 85 specimens of tumor tissues and adjacent tissues of gastric cancer patients from March 2016 to May 2018 were selected. The WISP2 expression in gastric cancer tissues and adjacent tissues was detected by immunohistochemistry. The WISP2-shRNA and NC-shRNA stable cell lines were constructed, and the effect of WISP2 down-regulation on migration of gastric cancer cells was analyzed by scratch wound healing assay. Transwell was used to detected the effect of WISP2 down-regulation on the invasion of gastric cancer cells. The effects of WISP2 down-regulation on the expression of E-cadherin, N-cadherin and Vimentin in gastric cancer cells were detected by western blotting.
