MicroRNA-132在结肠癌中的表达及其临床意义

目的  探讨Micro RNA-132（miR-132）在结肠癌中的表达及其临床意义。方法  实时逆转录定量PCR和原位杂交分析结肠癌组织中miR-132的表达情况。分析miR-132表达与结肠癌临床病理参数和预后之间的相关性。结果  miR-132在结肠癌组织中的表达明显低于正常结肠黏膜组织（P <0.01）,miR-132低表达与结肠癌分期更晚有关。多元回归分析表明,miR-132低表达与结肠癌患者生存期更短有关（P <0.01）,miR-132是结肠癌患者预后的独立预测因子。结论  miR-132表达下调能够促进结肠癌进展,是预测结肠癌患者预后的可靠指标,miR-132也具有成为结肠癌靶向治疗靶点的潜力。

Expression and roles of microRNA-132 in conlon carcinoma

Objective To investigate the association of miR-132 expression with clinicopathological characteristics and overall survival in patients with colon cancer. Methods Expression levels of the miR-132 were detected using qRT-PCR and in situ hybridization. Associations of miR-132 expression with clinicopathological factors and overall survival were statistically evaluated. Results The expression levels of miR-132 in the colon cancer tissues were significantly lower than those in the normal colon tissues, in line with the findings of in situ hybridization (P < 0.01). In addition, the tumors with low miR-132 expression were more likely to have advanced clinical stage (both P = 0.006) and higher grade (P = 0.01 and 0.02). Moreover, univariate analysis showed that the colon cancer patients with low miR-132 expression had shorter overall survival (P < 0.01). Multivariate statistical analysis further identified miR-132 as an independent prognostic factor for colon cancer (P < 0.01). Conclusions miR-132 low-expression may promote the aggressive tumor progression and be recognized as a reliable marker to predict the survival in patients with colon cancer. The three miRNAs could be attractive therapeutic targets in patients with advanced-stage colon cancer.
