摘    要:
目的:探讨淫羊藿苷对前列腺癌细胞株Du145与PC3的存活、迁移与侵袭能力的影响,并初步研究其机制。方法:CCK-8法检测不同浓度淫羊藿苷(0、5、10、20、40和80μmol/L)对Du145细胞与PC3细胞活力的影响;Transwell小室实验检测细胞的迁移和侵袭能力;Western blot检测Notch-1、MMP-2、MMP-9和Hes-1蛋白水平。结果:MTT法检测结果显示淫羊藿苷呈浓度依赖性抑制Du145与PC3细胞的活力,当浓度到达40μmol/L时,抑制作用达到最大;经淫羊藿苷干预后,Du145和PC3细胞迁移和侵袭能力显著下降,Notch-1、MMP-2、MMP-9和Hes-1蛋白表达水平显著下降。结论:淫羊藿苷具有抑制前列腺癌细胞株Du145与PC3生长、迁移和侵袭的作用,其作用机制可能与淫羊藿苷抑制Notch-1、MMP-2、MMP-9和Hes-1蛋白的表达有关。

Abstract:
AIM: To investigate the effects of icariin on the viability, migration and invasion of the prostate cancer lines Du145 and PC3.METHODS: Du145 cells and PC3 cells were treated with icariin at different concentrations (0, 5, 10, 20, 40 or 80 μmol/L), and the cell viability was measured by CCK-8 assay. The cell migration and invasion abilities were detected by Transwell assay. The protein expression of Notch-1, matrix metalloproteinase (MMP)-2, MMP-9 and hairy/enhancer of split-1 (Hes-1) was determined by Western blot.RESULTS: The results of MTT assay revealed that icariin inhibited the viabilitiy of Du145 cells and PC3 cells in a dose-dependent manner. The maximal effect was at dose of 40 μmol/L. Icariin treatment significantly decreased the abilities of migration and invasion of Du145 cells and PC-3 cells. Moreover, the protein expression of Notch-1, MMP-2, MMP-9 and Hes-1 was dramatically reduced after icariin treatment.CONCLUSION: Icariin inhibits prostate cancer cell viability, migration and invasion by decreasing the protein expression of Notch-1, MMP-2, MMP-9 and Hes-1.
