大鼠脊髓半切损伤模型的制备

摘    要:目的 用一种新的方法建立简便易行,稳定可靠的脊髓半切损伤模型.方法 取45只SD大鼠随机分为3组:假手术组为A组5只,40只为手术组(其中20只用眼用手术尖刀通过T11~T12椎间隙切割脊髓一侧1/2制成hSCI模型为B组,另20只用有齿血管钳咬除T11-12棘突和1/2椎板,用显微剪在T13~L1节段脊髓剪断其一侧1/2制成hSCI模型为C组).术后不同时间观察脊髓组织学变化,并记录脊髓神经功能综合评分(CBS).结果 光镜下伤侧灰质缩小,脊髓远端侧索和灰质神经变性坏死,出现大量空泡而神经元减少且明显萎缩,Waller's溃变下节段较上节段严重,术后CBS评分两两比较q检验发现手术组与假手术组有显著意义(P<0.01),但B组与C组CBS评分无显著性差异(P>0.05).结论 用眼用手术尖刀切割和显微剪切割制成hSCI模型具有同样的模型效果,但前者制备的模型死亡率低,更简单易行、安全可靠等优点.

Abstract:Objective To set up a practical and reliable model of hemi-sected spinal cord injury(hSCI)with a new and simple method.Methods 45 Srague-Daweley rats were divided at random into sham-operated group which was named group A including 5 rats and group B including 20 rats whose spinal cords between that of T_(13) and L_1 were hemisected by Ophthalmic sharp knife through the gap of T_(11)-T_(12),and group C whose spinal cords between that of T_(13) and L_1 were hemisected by micro-scissor after removing half lamina of vertebral arch and spinous process of T_(11) and T_(12) and displaying spinal cords of T_(13) and L_(1).At different times after injury.The histological change of lesioned cord was observed and evaluated under microscopy.Combined behavior scale(CBS) was employed for above 3 groups after operation every day,and difference among them was analyzed by q test.Results(It's) showed that the decrease of areas of gray matter in hSCI groups were observed being concomitant with a bath of empty cavities in gray matter in which neurons shrunk in lesion side.Moreover,Waller's degeneration in proximal segment was more serious than that in distal segment.In addition,the results showed that the difference of the CBS assessment in operated groups and sham-operated group at 24 hours,7 days and 21 days after operation was significant(P<0.01),however,the difference of CBS assessment of above group B and group C had no significance(P>0.05).Conclusions The models of hSCI which were maded ophthalmic sharp knife and micro-scissor respectively had the same effect of model.But the fomer model of hSCI had the advantages of low moritality.Besides,it is simple and stable.
