吡咯烷二硫氨基甲酸脂对缺氧再给氧冠状动脉内皮细胞凋亡的影响

摘    要:目的:探讨缺氧再给氧(H/R)时冠状动脉内皮细胞(CAEC)氧自由基清除功能和细胞凋亡率的变化及抗氧化剂吡咯烷二硫氨基甲酸脂(PDTC)对它的影响。方法:将体外培养的猪CAEC分为3组。对照组:未经处理;H/R组:作H/R处理;PDTC组:在H/R前于培养液中加入PDTC。分别检测各组细胞谷胱甘肽(GSH)、丙二醛(MDA)含量、核转录因子(NF-κB)表达及细胞凋亡率。结果:实验期对照组GSH、MDA含量、NF-κB的表达及细胞凋亡率无明显改变;H/R及PDTC组GSH含量明显低于对照组同期,而MDA含量、NF-κB表达及细胞凋亡率明显高于对照组同期;但PDTC组GSH含量明显高于H/R组同期,MDA含量、NF-κB的表达及细胞凋亡率明显低于H/R组同期。结论:H/R使细胞GSH含量明显减少,MDA含量、NF-κB的表达及细胞凋亡率明显增加;PDTC能有效降低NF-κB的表达,改善细胞氧自由基清除功能,降低细胞凋亡率,减轻H/R损伤。

Abstract:A Objective: To study the change of antioxidation function, and apoptosis of coronary artery endothelial cell during hypoxia/reoxygenation (H/R) and the effect of Pyrrolidine Dithiocar-bamate (PDTC) on it. Method:The culture porine coronary artery endothelial cells (CAECs) were divided into 3 groups, control group: porine CAECs not underwent treatment. H/R group: porine CAECs were exposed to hypoxia and then returned to reoxygenation condition. PDTC group: PDTC were added to the endothelial cells in the culture media before exposing to hypoxia condition. The endothelial content of GSH and MDA, the expression of NF-IcB were measured. Apoptosis rate was detected by flow cytometry. Result: At each time point, the endothelial content of GSH and MDA, the NF- κB,the Apoptosis rate of control group were not changed significantly. The cell content of GSH in H/R group and PDTC group was significantly lower than that in control group. Both levels of CAECs MDA, NF-κB and Apoptosis rate in H/R group and PDTC group was significantly higher than that in control group, but the cell content of GSH were significantly higher and the levels of CAECs MDA, NF-κB and Apoptosis rate were significantly lower in PDTC group than that in H/R group at each time point (P 〈 0.05). Conclusion: It seems that H/R decrease GSH and increace MDA, NF-κB espsessins rate of apoptosis in CAEC. PDTC can inhibite the expression of NF-kB, improve endothelial antioxidation function, reduce Apoptosis rate, and attenuate H/R injury.
