摘 要: | [目的]探讨活血燥湿汤对非酒精性脂肪肝大鼠的作用机制.[方法]以高脂肪、高胆固醇饲料喂养制作大鼠非酒精性脂肪肝模型.将30只SD大鼠随机分为对照组、模型组与中药干预组,每组10只,分别喂养16周后,测量动物的体重、肝重,常规苏木精-伊红染色观察肝组织病理变化,观察并比较对照组、模型组、干预组对肝组织羟脯氨酸(Hyp)、血清脂联素(APN)水平的影响.[结果]①病理学观察显示:模型组造模均成功,据肝细胞脂肪变性占所获肝组织标本量的范围分为4度(F0~F4),模型组炎症活动度积分(2.58±0.26),与对照组比较,差异有统计学意义(P<0.01);干预组炎症活动度积分(1.42±0.11),与模型组比较,差异有统计学意义(P<0.05).②模型组ALT、AST、TC、TG、Hyp含量均比对照组显著增高,模型组在ALT、AST、TC、Hyp含量上与干预组比较,差异有统计学意义;干预组ALT、AST、TC含量均比对照组增高,TG、Hyp含量与对照组比较,差异无统计学意义.③与对照组比较,模型组血清脂联素显著降低;干预组与模型组比较,血清脂联素差异有统计学意义;模型组及干预组肝组织脂联素含量与对照组比较均增高,差异有统计学意义.相关分析显示,血清脂联素水平与肝组织羟脯氨酸呈负相关.[结论]活血燥湿汤可能通过提高血清脂联素的含量、抑制肝组织羟脯氨酸的表达对非酒精性脂肪肝大鼠产生保护作用.

Abstract: | [Objective]To investigate the effect of huoxuezaoshi compound on rats with nonalcoholic fatty liver and its mechanism.[Methods] High-fat,high cholesterol diet were used to produce nonalcoholic fatty liver model,30 SD rats were randomly divided into normal group,model group and TCM intervention group,N =10,After 16 weeks of feeding,measuring the animal's weight,liver weight,HE staining was used to observe the liver histopathological changes; liver tissue hydroxyproline(Hyp),adiponectin(APN) levels were also measured in different groups.[Results]① Pathological observation showed that the inflammatory activity score in model group was 2.58±0.26,(P<0.01 vs control group);The inflammatory activity score in intervention group was 1.42±0.11,compared with model group,the difference was statistically significant (P<0.05).②In the model group,ALT,AST,TC,TG,Hyp content were significantly higher than control group;Compared with the control group,the difference in intervention group was not statistically significant.③Compared with the normal control group,,the level of serum adiponectin in model group was decreased significantly; compared with model group,the level of serum adiponectin in intervention group was up-regulated significantly; the level of adiponectin in liver tissue in model group and intervention group were higher than that in control group.Correlation analysis showed that serum adiponectin levels and liver hydroxyproline was negatively correlated.[Conclusion] Huoxuezaoshi compound could up-regulate the levels of serum adiponectin and inhibit the expression of liver hydroxyprolinein rats with nonalcoholic fatty liver. |
