补脾益气对脾虚证小鼠学习记忆的影响

目的:观察补脾益气中药对脾虚症小鼠学习记忆的影响,并探讨其作用机制。方法:采用利血平皮下注射建立脾虚小鼠模型,补脾益气中药治疗后观察小鼠的行为学改变,采用免疫组化法检测小鼠脑内神经元型一氧化氮合酶(nNOS)蛋白表达的变化。结果:脾虚证小鼠学习能力、记忆能力均下降;海马CA1区nNOS表达减少而大脑皮层表达增加。补脾益气中药能改善脾虚小鼠的学习能力,但对记忆能力影响不大;中药能调节小鼠海马CA1区及大脑皮层nNOS蛋白表达阳性细胞数(P<0.05,P<0.01)。结论:补脾益气中药可能通过调节nNOS蛋白来改善学习记忆能力。

Influence of Invigorating Spleen and Replenishing Qi Herbals on Learning and Memory of Mice with Spleen Deficiency

Objective: To observe the effect of invigorating spleen and replenishing qi herbals on learning and memory functions of mice with spleen deficiency and explore the underlying mechanism.Methods: The spleen-deficiency model was applied by injecting reserpine under subcutaneous dorsum.Alteration of behaviors after herbal treatment was observed in mice.The genetic expression of neuronal nitric oxide synthase(nNOS) in the brain tissue was determined by immunohistochemistry.Results: Compared with the normal group,the ability of learning and memory significantly reduced in the model group.After herbal treatment,the ability of learning was improved but the memory function did not;the expression of nNOS decreased in CA1 of hippocampi area but increased in cortex(P<0.05,P<0.01).Conclusion: Invigorating spleen and replenishing qi herbals can improve the function of learning by regulating the content of nNOS in the brain.
