摘    要:
目的探讨胰岛素对SD大鼠平滑肌细胞表达基质金属蛋白酶9(MMP-9)、组织抑制因子1(TIMP-1)以及内皮细胞表达可溶性E-选择素(s ES)的影响。方法原代培养SD大鼠颈内动脉平滑肌细胞及主动脉内皮细胞,用不同剂量的胰岛素作用后,以酶联免疫吸附实验(ELISA)分别测定上清液MMP-9、TIMP-1及s ES的水平。结果与对照组(无胰岛素)相比,胰岛素组MMP-9及s ES水平显著增高,TIMP-1则显著降低(P0.05)。经Pearson相关分析显示,胰岛素浓度与MMP-9(r=0.815,P0.001)及s ES(r=0.841,P0.001)呈正相关,与组织抑制因子1(r=-0.740,P0.001)呈负相关。结论胰岛素促进SD大鼠平滑肌细胞MMP-9及内皮细胞s ES表达,抑制平滑肌细胞TIMP-1表达,并呈剂量依赖性。
