淋球菌耐青霉素质粒转化大肠杆菌的研究

摘    要:
应用碱裂解法从淋球菌E参考菌株中获得约7.2kb耐青霉素质粒DNA,并转化大肠杆菌HB101内。采用DNA直接酶标——化学发光自显影技术,标记4.3kbpBR322质粒为探针,通过Southern印迹杂交加以鉴定,本实验所获含耐青霉素质粒的大肠杆菌HB101,将为淋球菌耐青霉素质粒探针的制备,检测耐青霉素基因打下基础。

Abstract:
About 7.2kb penicillin-resistant plasmid DNA was obtained from the reference strain-E of Neisse-ria gonorrhoeae,and was transfered to strain-HB101 of Escherichia Coli.Direct DNA labelling-chemolu-minescence photographic detection was used to label 4.3kb pBR322 plasmid probe.Transformation wasidentified by southern blotting hybridization,Strain-HB101 of Escherichia Coli obtained in the test whichcontained about 7.2kb penicillin-resistant plasmid DNA could be used to prepare the probe of penicillin-resistant plasmid of neisseria gonorrhoeae. The foundation was established for the exannnatjon of peni-cillin-resistant gene of Neisseria gonorrhoeae
