口腔白色念珠菌胶体金免疫层析试纸条的研制

目的：采用胶体金免疫层析技术研制快速检测口腔白色念珠菌（ Candida albicans）的试纸条。方法：用胶体金标记白色念珠菌单抗制备金标垫，制备白色念珠菌多抗，将白色念珠菌多抗和羊抗鼠IgG分别包被于NC膜上作为检测线和质控线，优化制作条件，组装成白色念珠菌胶体金免疫层析试纸条，并测定其特异性和灵敏度。结果：制备的白色念珠菌检测试纸条有良好的特异性，与口腔中常见的变形链球菌（ Streptococcus mutans）、粘性放线菌（Actinomyces viscosus）、血链球菌（Streptococcus sanguis）、干酪乳杆菌（Lactobacillus casei）、牙龈卟啉单胞菌（Porphy-romonas gingivalis）均无明显交叉反应，灵敏度为105 CFU／mL，检测耗时约5 min。结论：所制备的白色念珠菌胶体金检测试纸条具有特异性强、灵敏度高、操作简单、检测快速等优点，适用于临床上白色念珠菌的初步检测。

Development of a colloidal gold immunochromatographic test strip for detection of Candida albi-cans

Objective:To develop a colloidal gold immunochromatographic test strip for rapid and accu-rate detection of Candida albicans.Methods:The anti-C.albicans monoclonal antibody was conjugated with col-loidal golds and conjugate pad was prepared.The rabbit anti-C.albican polyclonal antibody was developed,and then tha rabbit anti-C.albican polyclonal antibody and goat anti-mouse IgG were separately dispensed on nitrocel-lulose membrane to form the test line( T) for sample detection and the control line( C) for quality-control,respec-tively.After the condition parameters were optimized,the gold immunochromatography test strip for C.albicans detection was assembled,and specificity and sensitivity were determined.Results:The prepared C.albicans test strip had a good specificity,no cross-reaction with other five common bacteria usually existed in the mouth,such as Streptococcus mutans,Actinomyces viscosus,Streptococcus sanguis,Lactobacillus casei and Porphyromonas gingi-valis Aeromonas were found.The detection sensitivity was 105 cfu/mL of C.albicans.The positive results could be easily judged by the presence of a red test line with naked eye within 5 min.Conclusion:The developed test strip is specific,simple and convenient for rapid detection of C.albicans presenting good specificity and high sensitivi-ty,.It will become a useful technique for the initial detection of C.albicans in clinic.
