卵巢过度刺激综合征大鼠模型建立

摘    要:
为建立卵巢过度刺激综合征大鼠模型 ,将 2 2日龄大鼠 1 8只随机分成卵巢过度刺激组、超排卵组和生理盐水对照组 ,每组 6只。按照改良的 Ujioka方法 ,卵巢过度刺激组从第2 2日龄起皮下注射 PMSG1 0 IU连续 4d,第 2 6日龄注射 HCG1 0 0 IU;超排卵组从第 2 4日龄皮下注射 PMSG 1 0 IU,第 2 6日龄注射 HCG 1 0 IU,其余时间同上注射生理盐水 ;生理盐水对照组经皮下注射生理盐水 0 .1 ml连续 5d即从第 2 2~ 2 6日龄。在 HCG注射 72h后 ,尾静脉注射 1 % Evans blue( EB) 0 .1 ml以评价腹腔毛细血管通透性。EB注射 3 0 min后 ,观察卵巢重量、腹腔 EB含量及有无腹水出现。结果表明 :卵巢过度刺激组卵巢重量 ( 0 .3 53± 0 .0 2 2 8g)显著高于超排卵组和对照组卵巢重量 ( 0 .1 3 5± 0 .0 2 8g,0 .0 57± 0 .0 1 0 ;分别 P<0 .0 0 1 ) ;卵巢过度刺激组腹腔 EB含量 ( 2 .3 3± 1 .0 6μg)显著高于超排卵组和对照组卵巢重量 ( 0 .79± 0 .1 8μg,0 .68± 0 .1 9μg;分别 P<0 .0 0 1 )。卵巢过度刺激组出现腹水 ,而超排卵组和生理盐水对照组无腹水出现。这提示 :卵巢过度刺激综合征大鼠模型建立是成功的 ,该模型为系统性探讨卵巢过度刺激综合征的确切发病机理奠定了一定的基础。

Abstract:
To establish a rat model of ovarian hyperstimulation syndrome, a total of 18 immature female rats were randomly divided into three groups: ovarian hyperstimulation group, superovulation group and control group. According to our modification of Ujioka method, six rats in ovarian hyperstimulation group were given a subcutaneous injection of 10 IU pregnant mare serum gonadotropin (PMSG) in 0.1ml 0.9% saline for 4 consecutive days, then given 100 IU human chorionic gonadotropin (HCG) in 0.1 ml 0.9% saline on the fifth day (26th day of life).In superovulation group, six rats were given 10 IU PMSG at the time indicated above on the 24th day of life, and were given 10 IU HCG 48 h later (26th day of life).In control group, the rats were given the same dosage of 0.9% saline for 4 consecutive days, from the 22nd day to 26th day of life. 72 h after HCG injection, 0.1 ml 1% EB was injected into each rat by tail vein in order to determine the peritoneal capillary permeability. 30 min after the last injection, we found that ovarian weight, peritoneal EB level and ascitic score of ovarian hyperstimulation group were significantly higher than those of superovulation group and control group (P<0.001 respectively).These results demonstrated that the establishment of rat model of ovarian hyperstimulation syndrome was successful and may be useful for research on the pathogenesis of the ovarian hyperstimulation syndrome in the future.
