摘 要: | 目的 MMR系统是一组高度保守基因及其产物蛋白构成,其主要功能是纠正DNA复制、重组及基因损伤时出现的碱基配对错误.本研究总结国内外错配修复基因与大肠癌相关性的研究进展.方法 检索Pubmed和万方数据库,以“错配修复基因、微卫星不稳定、大肠癌”等为关键词,检索1997-01-2016-06的相关文献共102篇.纳入标准:(1)错配修复基因与微卫星不稳定的关系.(2)错配修复基因大肠癌的发生及预后的关系.根据纳入标准,最终选取49篇文献纳入分析.结果 错配修复系统是一种广泛存在于机体细胞内的自我修复机制,对防止基因突变和维持基因组稳定性方面起着重要作用.错配修复基因突变与微卫星不稳定关系密切,错配修复基因发生突变时,使机体肿瘤易感性明显增加.结论 错配修复系统是大肠癌的发生、发展的重要预后指标.

Abstract: | OBJECTIVE To sum up the status of correlation between colorectal cancer and mismatched repair system(MMR) which includes a group of highly conserved genes and protein as the products of genes to correct base pairing mistakes while DNA is replicating,recombining or damaged.METHODS Totally 102 relevant articles were searched with "mismatched repair system(MMR),microsatellite instability(MSI),colorectal cancer(CRC)" as keywords in PubMed and WanFang database from January 1997 to June 2016.Totally 49 papers were selected and analyzed according to the inclusion criteria as follows:relationship between MMR and MSI;relationship between MMR and CRC in oncogenesis and prognosis.RESULTS Mismatched repair system represents a self-repaired mechanism existing in human cells,which plays an important role in preventing genetic mutations and maintaining genetic stability.Mismatch repair gene mutation and microsatellite instability are closely related.Mismatched repair gene mutations lead to the increaseof tumor susceptibility.CONCLUSION The status of MMR was important prognostic indicator of tumor genesis and growth of CRC. |
