Abstract:
The present paper was designed to observe the mitotic(spermatogonia) and meiotic(primary and secondary spermatocytes) chromosomes prepared directly from spermatogenic cells in semen ejaculates collected at different period of intervals of healthy and fertile men by treatment with hypotonic saline and sodium citrate solution, The results obtained are as follows,(1) Majority of the mitotic chromosomes from the exfoliated spermatogonia of normal humen semen treated with hypotonic solution were shown to be shorter and thicker with deep stained metaphase diploid chromosomes. While the chromosomes of weiotic division from primary and secondary spermatocytes were mostly of longer and thinner ones of haploid or diploid chromatids,(2) By these procedures, more prophase I figures including stages of zygotene that with synaptonemal complex, pachytene with tetrad,diplotene with chiasma and segregation of sister chromatids, and diakinesis with apparent chromosome condensation can be obtained,less figures of anaphase I,telophase I and the secondary meiotic division were seen. Semens from two ejaculates at interval of less than 24 hrs. were not readily to prepare spermatogenic cell chromocomes.The longer intervals between ejaculations, the more division figures could be obtained.(3) It was shown that tail-like fibril structyre was observable at the long arm(q) terminal of sex chromosomes in prophase I(but never found in metaphase) of spermatogenic cells.(4) By these procedures more metaphase, figures could be obtained,the metaphase index was 11.5%,in which the majority of normal human spermatogenic cells were haploid and theratio of diploid v. s haploid was 1:2(222/446).However triploid(0.58%, 4/689)and tetraploid(2.5%,17/689) chromosomes were frequently seen, which might be caused by nondisjunction during meiosis, the structure of post prophase were never seen.
